Summary: Recent progress in the isolation and characterization of HIV-specific broadly neutralizing antibodies has paved the way for vaccine development and exploring passive administration of monoclonal antibodies for HIV prevention. While AMP trials with VRC01 showed promising results in preventing HIV acquisition, further research on newer, more potent bnAbs is underway to improve overall prevention efficacy. Iterative phase 1 trials with dual and triple bnAbs, as well as bi- and trispecific antibodies, are being conducted to build on the findings of the VRC01 efficacy trials and enhance prevention strategies for HIV.

Development of a VRC01-class germline targeting immunogen derived from anti-idiotypic antibodies

Summary: Our study focuses on developing anti-idiotypic monoclonal antibodies (ai-mAbs) against inferred-germline (iGL) VRC01-class antibodies to selectively activate VRC01-class precursors. By engineering a bispecific molecule derived from two ai-mAbs, we were able to preferentially activate iGL-VRC01 B cells in vitro and induce specific antibody responses in a murine adoptive transfer model with diverse polyclonal B cells. This molecule represents an alternative non-envelope-derived germline-targeting immunogen that shows promising results in selectively activating VRC01-class precursors in vivo.
