Association of Systemic Inflammatory Factors with Progression to Advanced Age-related Macular Degeneration

Brandie D. Wagner et al.

Summary: Systemic inflammation may be associated with future disease progression in patients with intermediate AMD. Combinations of biomarkers can distinguish between patients who do and do not progress to advanced AMD, indicating a complex relationship rather than a simple increase in a few markers. Studying systemic inflammation in iAMD may provide insights into early pathologic events and potentially identify patients at highest risk for developing severe AMD.

Neurodegeneration, gliosis, and resolution of haemorrhage in neovascular age-related macular degeneration, a clinicopathologic correlation

Miaoling Li et al.

Summary: This study provides a comprehensive analysis of neurodegeneration and gliosis in patients with neovascular age-related macular degeneration, highlighting the importance of OCT imaging in understanding the pathophysiology of macular diseases. The findings suggest that intensified OCT reflectivity of the external limiting membrane may serve as a potential biomarker for severe photoreceptor loss and gliosis in these patients.

New Omics-Derived Perspectives on Retinal Dystrophies: Could Ion Channels-Encoding or Related Genes Act as Modifier of Pathological Phenotype?

Luigi Donato et al.

Summary: Ion channels, integral proteins expressed in various organs including the eye, play crucial roles in physiological processes like signal transmission and visual processing. Mutations in these channels and their interacting genes lead to a range of ocular diseases known as channelopathies. By conducting whole exome sequencing experiments on families with inherited retinal dystrophies, researchers found variants in causative genes affecting retinal functionalities. Further investigation revealed mutated genes interacting with neurotransmission pathways, offering new insights for precise diagnosis and treatment of retinal dystrophies.

Retinal Inflammation, Cell Death and Inherited Retinal Dystrophies

Lorena Olivares-Gonzalez et al.

Summary: Inherited retinal dystrophies (IRDs) are a group of retinal disorders that lead to progressive vision loss due to retinal cell death, predominantly photoreceptor cells. The treatment of IRDs is complicated by genetic and clinical heterogeneity, but reducing retinal inflammation has emerged as an effective approach to slowing disease progression.

Optimized Adeno-Associated Virus Vectors for Efficient Transduction of Human Retinal Organoids

Manuela Voelkner et al.

Summary: rAAV vectors are widely used in gene delivery in the retina, showing safety and efficacy in treating inherited retinal dystrophies with limitations in transduction efficiency. Novel variants with modified capsid sequences have been engineered to improve this efficiency. Testing on hiPSC-derived human retinal organoids showed promising results with AAV9.NN achieving the highest efficiency in transduction. Using human organoids provides a valuable platform for evaluating novel AAV vectors for gene therapy applications in the retina.

Age-related macular degeneration

Monika Fleckenstein et al.

Summary: Age-related macular degeneration (AMD) is the leading cause of legal blindness in the industrialized world, with a complex pathophysiology involving aging, environmental factors, and genetic susceptibility leading to photoreceptor degeneration and drusen formation. Tailoring therapeutics to specific phenotypes and stages may be crucial for preventing irreversible vision loss in the future.

Summary: The optimized protocol for high yield mouse retinal organoids facilitates the assessment of key parameters, showing reliable completion of retinogenesis and maintenance for at least 60 days in culture with continued maturation. Activation of sonic hedgehog signaling during development increased retinal epithelial length in the organoid system, supporting organoid maturation, stability, and reproducibility.

Implication of specific retinal cell-type involvement and gene expression changes in AMD progression using integrative analysis of single-cell and bulk RNA-seq profiling

Yafei Lyu et al.

Summary: The study utilized single-cell RNA sequencing to analyze gene expression in retinal cells of AMD patients, revealing changes in cell types and gene expression in AMD eyes compared to normal eyes, particularly in certain specific cell types in the macula region. KEGG pathway analysis showed enrichment in pathways related to complement, coagulation, antigen presentation, and signaling in AMD eyes, highlighting part of the pathogenesis of AMD.
