Comprehensive insulin receptor phosphorylation dynamics profiled by mass spectrometry

Zhongping Liao et al.

Summary: In this study, an extensive assessment of IR phosphorylation was conducted using nano-liquid chromatography-tandem mass spectrometry, revealing diverse phosphorylation kinetics across different sites in response to different receptor agonists. The research also found that an IR peptide agonist was unable to induce IR trafficking to early endosomes, likely due to differences in IR phosphorylation. This study provides a powerful tool for investigating IR signaling and trafficking, which can aid in designing IR agonists with improved therapeutic efficacy.

Functionally selective signaling and broad metabolic benefits by novel insulin receptor partial agonists

Margaret Wu et al.

Summary: This study characterizes insulin dimers that act as insulin receptor partial agonists, exhibiting hepatic and adipose tissue preference and metabolic benefits. The partial agonism is achieved through distinct conformational changes of the insulin receptor and specific binding properties of the dimers. The tissue-selective action of these insulin receptor partial agonists could offer a potentially safer approach for diabetes treatment.

An aptamer agonist of the insulin receptor acts as a positive or negative allosteric modulator, depending on its concentration

Na-Oh Yunn et al.

Summary: Researchers have discovered a unique aptamer modulator called IR-A62 that affects the interaction between insulin and its receptor protein. The action of IR-A62 depends on its concentration relative to insulin, with low concentrations enhancing insulin binding and high concentrations competing with insulin for the receptor. In addition, injecting IR-A62 can lower blood glucose levels in diabetic mice with a longer-lasting effect than insulin injection. These findings suggest that aptamers can elicit various responses from receptors, expanding the potential therapeutic applications of aptamers.

A hotspot for enhancing insulin receptor activation revealed by a conformation-specific allosteric aptamer

Na-Oh Yunn et al.

Summary: Aptamers are single-stranded oligonucleotides that can bind to a specific target with high affinity. In this study, researchers discovered that aptamers can also act as positive allosteric modulators to enhance receptor activation. By developing a novel aptamer, IR-A43, they demonstrated that it could potentiate insulin receptor activation by binding to the receptor and stabilizing ligand binding.
