DIVERSITY AND BIOLOGY OF CANCER-ASSOCIATED FIBROBLASTS

Giulia Biffi et al.

Summary: Efforts to develop anti-cancer therapies have mainly targeted the epithelial compartment, but recent studies have shown the significant influence of cancer-associated fibroblasts (CAFs) in tumor progression. CAFs not only promote cancer cell proliferation, therapy resistance, and immune exclusion, but may also restrain tumor progression in certain contexts. Research on CAFs has focused on their heterogeneity, plasticity, and functions across different cancer types and states, as well as advancements in therapeutic strategies targeting CAFs currently undergoing preclinical and clinical evaluation.

Liquid biopsy enters the clinic - implementation issues and future challenges

Michail Ignatiadis et al.

Summary: The approval of liquid biopsy assays for detecting genetic alterations in plasma cell-free DNA has marked a tipping point for the clinical use of this technology, especially in patients with advanced-stage cancer. The next frontier is likely to be the systemic treatment of patients with 'ctDNA relapse' and other potential future applications include cancer screening and diagnosis.

DUSP16 promotes cancer chemoresistance through regulation of mitochondria-mediated cell death

Heng Boon Low et al.

Summary: The study identifies DUSP16 as a regulator of chemoresistance in various cancers by impairing mitochondria-associated apoptosis. DUSP16 is proposed as a marker for chemotherapy sensitivity and treatment outcome, offering a potential therapeutic target for overcoming chemoresistance in cancer therapy.
