The role of NSP6 in the biogenesis of the SARS-CoV-2 replication organelle

Summary: The replication organelle of SARS-CoV-2 is generated by viral non-structural proteins (NSPs) NSP3 and NSP4, while NSP6 plays a role in connecting ER membranes and organizing DMV clusters. NSP6 also mediates the contact between SARS-CoV-2 and lipid droplets (LDs), providing a means to refurbish the replication organelle with LD-derived lipids.

Summary: This study investigated the cell-cell fusion induced by the Old World Lassa virus GPC complex and found that viral entry into host cells is mainly regulated by the human LAMP1 receptor and a specific anionic lipid, bis(monoacylglycero)phosphate. The findings suggest that arenaviruses have evolved to utilize bis(monoacylglycero)phosphate to enter cells from late endosomes.

Dependence of SARS-CoV-2 infection on cholesterol-rich lipid raft and endosomal acidification

Summary: This study found that SARS-CoV-2 enters human cells through ACE2 and is sensitive to the pH of endosomes/lysosomes in HEK293T cells, with infection dependent on cholesterol-rich lipid rafts instead of other pathways. These findings expand the understanding of the entry mechanism of SARS-CoV-2 and suggest a new strategy for combating the virus.
