Multi-Omic Analysis to Characterize Metabolic Adaptation of the E. coli Lipidome in Response to Environmental Stress

Summary: In response to external stress, bacteria undergo dynamic remodelling of lipid metabolism pathways to alter cellular membrane composition. Using Escherichia coli as a model system, a 'multi-omics' strategy was developed for quantitative analysis of lipidome and proteome profiles during exponential versus stationary growth phases. Analysis revealed a decrease in total lipid content and increase in cardiolipin composition and odd-numbered acyl-chain glycerophospholipids under stationary growth conditions. Field asymmetry ion mobility spectrometry and ultraviolet photodissociation-tandem mass spectrometry enhanced the depth of coverage and lipid structural characterisation. Proteome analysis identified key enzymes involved in adaptive lipid metabolism, phospholipid metabolism, fatty acid synthesis and degradation, and oxidative stress response in E. coli.
