The relief of hypoxic microenvironment using an O2 self-sufficient fluorinated nanoplatform for enhanced photodynamic eradication of bacterial biofilms

Lingyun Zou et al.

Summary: The study developed oxygen-sufficient fluorinated liposomes that enhanced the photodynamic eradication of bacterial biofilms by relieving the hypoxic microenvironment and promoting ROS generation under laser irradiation. The co-delivered O-2 carrier PFH significantly improved the bactericidal capability of PDT. Lip-Ce6-PFH@O-2 showed better antibiofilm ability than Lip-Ce6, alleviated inflammation symptoms, and accelerated wound healing in a mouse model, providing a new paradigm for enhancing the therapeutic efficacy of PDT for efficient biofilm eradication.

An Ethacrynic Acid-Brominated BODIPY Photosensitizer (EA-BPS) Construct Enhances the Lethality of Reactive Oxygen Species in Hypoxic Tumor-Targeted Photodynamic Therapy

Miae Won et al.

Summary: This study introduces a new construct, EA-BPS, which induces a synergistic antitumor effect through a combination of ROS production and GST-π inhibition, leading to enhanced cell-killing effect under hypoxic conditions. By making better use of available oxygen in tumor environments, improved therapeutic PDT outcomes can be achieved even in low oxygen conditions.

Synthesis and Bioactivity of Guanidinium-Functionalized Pillar[5]arene as a Biofilm Disruptor

Shuwen Guo et al.

Summary: A novel guanidinium-functionalized pillar[5]arene, GP5, was designed and synthesized to effectively combat bacterial biofilms. GP5 showed high antibacterial potency against Gram-negative and Gram-positive strains and successfully disrupted preformed biofilms by penetrating biofilm barriers and destroying biofilm-enclosed bacteria. Additionally, host-guest complexation between GP5 and cefazolin sodium exhibited synergistic disruption activity against biofilms, providing a novel supramolecular platform for biofilm eradication.

BODIPY-Based Photodynamic Agents for Exclusively Generating Superoxide Radical over Singlet Oxygen

Kun-Xu Teng et al.

Summary: Developing Type-I photosensitizers that can generate superoxide radicals under light irradiation has been shown to be an effective approach for improving photodynamic therapy for hypoxic tumors. The reported alpha,beta-linked BODIPY dimers and trimer exclusively generate superoxide radicals through an efficient excited-state relaxation process, demonstrating superior performance in PDT under hypoxic conditions.

Boosting the inactivation of bacterial biofilms by photodynamic targeting of matrix structures with Thioflavin T

Patricia Bondia et al.

Summary: Thioflavin T (ThT), commonly used as a fluorescent probe for amyloid detection, has been discovered to exhibit photodynamic activity against bacterial biofilms. It disrupts the complex architecture of biofilm matrix and efficiently inactivates bacterial cells, while also synergistically enhancing the activity of conventional antimicrobials.
