Regulating T-cell differentiation through the polyamine spermidine

Guilhermina M. Carriche et al.

Summary: The study demonstrates that spermidine can act as a modulator of T-cell differentiation and function, promoting the development of Treg cells. Spermidine affects the differentiation of naive T cells both in vitro and in vivo, exerting beneficial effects on gut immunity.

Spermidine ameliorates high-fat diet-induced hepatic steatosis and adipose tissue inflammation in preexisting obese mice

Lingyan Ma et al.

Summary: Spermidine administration can reduce fat mass and plasma lipid profile in HFD-induced obese mice, improve hepatic steatosis, reduce adipose tissue inflammation, and enhance gut barrier function. Additionally, spermidine treatment also enhances thermogenic gene expression in brown adipose tissue.

Metabolic regulation by PD-1 signaling promotes long-lived quiescent CD8 T cell memory in mice

Vandana Kalia et al.

Summary: The inhibitory role of PD-1 signaling in CD8 T cells has been well established in chronic viral infections and cancers, but its impact on the development of long-lived antigen-independent T cell memory remains unclear. Studies have shown that PD-1 expression on antigen-specific CD8 T cells is essential for the formation of a durable CD8 T cell memory pool, with loss of T cell-specific PD-1 signaling leading to increased apoptosis and a defect in antigen-independent renewal of memory CD8 T cells.

eIF5A hypusination, boosted by dietary spermidine, protects from premature brain aging and mitochondrial dysfunction

YongTian Liang et al.

Summary: Mitochondrial function declines with age in the brain, and supplementing spermidine levels can delay this decline. The levels of hypusinated eIF5A decline with age in the Drosophila brain, but can be boosted by dietary spermidine. Attenuating eIF5A hypusination affects brain mitochondrial respiration and accelerates aging in locomotion and memory formation in adult Drosophilae, indicating it may be a potential diagnostic and therapeutic avenue for age-induced cognitive decline and neurodegeneration.

Polyamine metabolism is a central determinant of helper T cell lineage fidelity

Daniel J. Puleston et al.

Summary: Polyamine synthesis plays a critical role in T cell activation and differentiation, regulating CD4(+) T cells' function and epigenetic remodeling. Studies have revealed the significance of polyamine metabolism in controlling the fate of T-H cell subsets, with deficiency leading to aberrant expression of cytokines and transcription factors.

Critical role of the CD44low CD62Llow CD8+ T cell subset in restorinng antitumor immunnity in aged mice

Yuka Nakajima et al.

Summary: CD8(+) T cells play a crucial role in antitumor immune responses, but this function is suppressed in aged individuals. The mechanism of this age-related immune decline is unclear. This study found that inefficient generation of a specific CD8(+) T cell subset may contribute to age-related resistance to PD-1 blockade, which can be overcome by strong TCR stimulation.
