The Role of Oxidative Stress in the Pathogenesis of Vitiligo: A Culprit for Melanocyte Death

Yijie Xuan et al.

Summary: This review summarizes the role of oxidative stress in the pathogenesis of vitiligo and discusses the mechanisms of interaction between oxidative stress and various modes of cell death. The findings suggest that oxidative stress acts as an initiator for different cell death modes through complex mechanisms, such as autophagy, apoptosis, and ferroptosis. These insights provide new ideas for exploring the pathogenesis and potential therapeutic targets of vitiligo.

The role of HMGB1 in inflammatory skin diseases

Takashi K. Satoh

Summary: HMGB1 is a highly abundant non-histone nuclear protein that can promote the pathogenesis of inflammatory diseases. It can translocate to the cytoplasm and extracellular space through release and modifications and induce cytokine production or tissue repair through receptor binding. It can also form complexes with cytokines and enhance their biological effects. Cell death is a significant source of HMGB1, and different forms of cell death can modulate its activity. Blocking HMGB1 can attenuate disease severity in skin diseases. Therefore, HMGB1 may serve as a biomarker and therapeutic target in inflammatory skin diseases.

Does oxidative stress correlate with disease activity and severity in vitiligo? An analytical study

Sinu Rose Mathachan et al.

Summary: Oxidative stress likely plays a pathogenic role in vitiligo, with significantly raised levels of oxidative damage marker and decreased levels of antioxidants observed in patients. Generalized and active disease groups showed more derangement in oxidative stress parameters compared to localized and stable disease groups.

Elevated serum level of interleukin-15 in vitiligo patients and its correlation with disease severity but not activity

Mona A. Atwa et al.

Summary: Background Vitiligo is a common acquired disorder of depigmentation. IL-15, a cytokine in the IL-2 family, plays a crucial role in autoimmune diseases but has been poorly investigated in patients with vitiligo. The study found that serum IL-15 levels were significantly higher in patients with vitiligo compared to healthy controls, and there was a positive correlation between IL-15 levels and vitiligo severity.

Abu Saleh Md Moin et al.

Summary: Type 1 diabetes is a T-cell mediated autoimmune disease characterized by the recognition of pancreatic beta-cell proteins as self-antigens, leading to their loss. Heat shock proteins play diverse roles in cellular functions, protecting cells from stress and potentially impacting the pathology of diabetes.

Summary: Purinergic signaling is a crucial cell communication pathway mediated by extracellular nucleotides and nucleosides. Activation of P2 receptors influences the redox balance of cells, while enzymes like E-NTPDase1/CD39 and ecto-5'-nucleotidase/CD73 play key roles in regulating this process.

Post-Translational Modification of HMGB1 Disulfide Bonds in Stimulating and Inhibiting Inflammation

Ulf Andersson et al.

Summary: HMGB1 is a highly conserved nuclear DNA-binding protein that acts as a damage-associated molecular pattern molecule (DAMP) with oxidation-reduction sensitive properties. It can modulate proinflammatory and anti-inflammatory activities by binding to different cell surface receptors. HMGB1 plays a crucial role in innate immunity by carrying other DAMPs and PAMPs, leading to the induction of inflammation responses.

Summary: PEI induces immune responses by stimulating ATP release, activating VDAC-1 and pannexin 1 pathways, and increasing intracellular Ca2+ concentration. In vivo experiments show that PEI exposure leads to Th2 cytokine secretion and eosinophil recruitment, with IL-33 playing a crucial role in this process.

Summary: Bacterial infections caused by 'superbugs' are on the rise globally, with traditional antibiotics becoming less effective, posing a risk of entering a post-antibiotic era. Antimicrobial peptides (AMPs) have garnered attention for their clinical potential as a new class of antibiotics to combat antimicrobial resistance. This review discusses various aspects of AMPs, including their physicochemical properties, mechanisms of action, and chemical synthesis strategies for developing novel AMPs.
