Development of a pentavalent broadly protective nucleoside-modified mRNA vaccine against influenza B viruses

Norbert Pardi et al.

Summary: This study focuses on the development of a universal influenza B virus vaccine using the lipid nanoparticle-encapsulated nucleoside-modified mRNA (mRNA-LNP) platform. Vaccine candidates based on different target antigens are evaluated for their ability to protect against ancestral and recent influenza B viruses. The findings show that a pentavalent vaccine with multiple antigens can protect mice from morbidity even at a very low dose.

A chimeric hemagglutinin-based universal influenza virus vaccine approach induces broad and long-lasting immunity in a randomized, placebo-controlled phase I trial

Raffael Nachbagauer et al.

Summary: New influenza virus vaccines based on chimeric hemagglutinin have been found to induce a broad, strong, durable and functional immune response targeting the conserved stalk of the hemagglutinin protein. This suggests that these chimeric hemagglutinins have the potential to be developed as universal vaccines that protect broadly against influenza viruses.

mRNA vaccination induces tick resistance and prevents transmission of the Lyme disease agent

Andaleeb Sajid et al.

Summary: The study showed that vaccination with 19ISP mRNA can enhance host recognition of tick bites, reduce tick engorgement, and prevent B. burgdorferi infection. Immunization with 19ISP also hindered the transmission of the pathogen in vaccinated guinea pigs.

Antigen modifications improve nucleoside-modified mRNA-based influenza virus vaccines in mice

Alec W. Freyn et al.

Summary: The study focused on protein modifications to enhance the safety and efficacy of mRNA-based influenza vaccines. Findings showed that membrane-bound HA constructs elicited stronger antibody responses, altering the catalytic site of NA reduced reactogenicity while maintaining protective immunity, and NP was superior to M1 in conferring protection from influenza virus.
