Summary: In Gram-negative bacteria, approximately half of lipoproteins destined for the outer membrane display an intrinsically disordered linker at their N terminus. The unstructured linkers are required for optimal trafficking by the Lol lipoprotein sorting system, re-routing lipoproteins to the inner membrane without them. Replacing the linker with an artificial peptide of similar length and disorder can restore normal outer-membrane targeting in the stress sensor RcsF.

Mechanism of LolCDE as a molecular extruder of bacterial triacylated lipoproteins

The study reveals the mechanism of how LolCDE recognizes its substrate and the key role of the amide-linked acyl chain in this process. Nucleotide binding triggers large-scale, asymmetric movements in the structure of LolCDE, leading to extrusion of the captured lipoprotein. Comparisons with MacB highlight the conserved mechanism of type VII ABC transporters and the unique properties of LolCDE as a molecule extruder of triacylated lipoproteins.

The surface lipoproteins of gram-negative bacteria: Protectors and foragers in harsh environments

As more bacterial genomes are sequenced, the prevalence and different roles of SLPs are being discovered, offering potential as antimicrobial targets. Through understanding the structures and mechanisms of SLPs, alongside bioinformatics and genome sequencing, we may develop new therapies and vaccine antigens.
