Transcriptional heterogeneity and tightly regulated changes in gene expression during Plasmodium berghei sporozoite development

Summary: This study used single-cell RNA sequencing to characterize the gene expression profiles of Plasmodium berghei sporozoites throughout their development in mosquitoes. The results revealed tightly regulated changes in gene expression and highlighted candidate genes involved in various functions. Additionally, the study showed transcriptional heterogeneity among parasites and suggested that Plasmodium development in mosquitoes is asynchronous and regulated by intrinsic and environmental factors. Finally, the analysis demonstrated a decrease in transcriptional activity followed by rapid reactivation upon salivation in mature sporozoites.

The RNA structurome in the asexual blood stages of malaria pathogen plasmodium falciparum

Summary: In this study, the differential accumulation of RNA secondary structures in coding and non-coding transcripts from the asexual developmental cycle in P. falciparum was measured. Approximately 71% of genes with secondary structures were found to be protein coding mRNAs, with new structural RNA regions identified in the malaria genome. The mapping data corresponded to all regions of mRNAs and revealed stage-specific, dynamic relationships between mRNA structural contents and translation efficiency.
