Industry Perspective on the use and Characterization of Polysorbates for Biopharmaceutical Products Part 1: Survey Report on Current State and Common Practices for Handling and Control of Polysorbates

Klaus Wuchner et al.

Summary: This study report presents the industry practices, stability, and regulatory aspects of polysorbates in biopharmaceutical products, comparing the findings with available scientific literature. The upcoming second part of the report will focus on exploring the degradation pathways of polysorbates and developing effective control strategies.

Mechanistic understanding of metal-catalyzed oxidation of polysorbate 80 and monoclonal antibody in biotherapeutic formulations

Fethi Bensaid et al.

Summary: This study evaluated the oxidation of polysorbate 80 (PS80) and a monoclonal antibody (mAb) under different conditions. The presence of iron accelerated the oxidation of PS80 and increased the oxidation of a specific solvent-exposed mAb methionine. PS80-AO was more sensitive to oxidative degradation than PS80-MC. The kinetics of PS80 oxidation was pH-dependent in the presence of histidine buffer. Increasing mAb concentration exhibited a protective effect against metal catalyzed PS80 and methionine oxidation.

Complex Micellization Behavior of the Polysorbates Tween 20 and Tween 80

Hannah Knoch et al.

Summary: Polysorbates are versatile surfactants used for stabilizing biologicals and solubilizing active ingredients. This study characterizes the complex association behavior of two types of Polysorbates, showing that different Polysorbate species gradually enter initial micelles with increasing concentrations. The variation of micelle properties differs between Polysorbate grades and can progress up to at least 10 mM concentration.
