Downregulation of HULC Induces Ferroptosis in Hepatocellular Carcinoma via Targeting of the miR-3200-5p/ATF4 Axis

Lulu Guan et al.

Summary: This study found that knockdown of HULC increases ferroptosis and oxidative stress in liver cancer cells. They also identified changes in certain miRNAs in cells treated with HULC siRNA, with HULC acting as a ceRNA of miR-3200-5p. miR-3200-5p regulates ferroptosis by targeting ATF4, leading to the inhibition of proliferation and metastasis in HCC cells.

Sevoflurane Improves Hemorrhagic Shock and Resuscitation-Induced Cognitive Impairments and Mitochondrial Dysfunctions through SIRT1-Mediated Autophagy

Jianwei Shu et al.

Summary: Sevoflurane postconditioning (SP) can effectively attenuate spatial learning and memory impairments caused by cerebral ischemia reperfusion injury (IRI). IRI leads to reduced SIRT1 levels, accompanied by oxidative stress, mitochondrial dysfunction, and inactivated autophagy. However, SP or genetically upregulating SIRT1 can suppress defective autophagy, mitochondrial oxidative injury, and neuronal death. This study reveals the important role of SIRT1 in cerebral IRI.

The Transcription Factor p63 Is a Direct Effector of IL-4-and IL-13-Mediated Repression of Keratinocyte Differentiation

Anne M. Brauweiler et al.

Summary: Atopic Dermatitis is linked to defects in skin barrier function caused by increased levels of type-2 cytokines, repressing keratinocyte differentiation. Gene repression induced by type-2 cytokines requires the master regulator p63, affecting multiple genes involved in KC differentiation through pathways converging on transcription factors p63 and Notch.

Yahui Ding et al.

Summary: This study identified a novel approach to treat TNBC by inducing ferroptosis through ubiquitination of GPX4 with the compound DMOCPTL, and revealed the role of GPX4 in regulating mitochondria-mediated apoptosis through up-regulation of EGR1 in TNBC cells. Furthermore, the prodrug Compound 13 showed promising anti-tumor effects in vivo with no apparent toxicity, suggesting its potential as a lead compound for developing effective anti-TNBC drugs.

Arsenite induces ferroptosis in the neuronal cells via activation of ferritinophagy

Junting Xiao et al.

Summary: This study demonstrated that arsenite can trigger ferroptosis in neuronal cells through activation of ferritinophagy. Regulating ferritinophagy to control iron levels may provide a clue for prevention against arsenite neurotoxicity. Co-treatment with ferrostatin-1 or autophagy inhibitors significantly attenuated the cytotoxic effects of arsenite.

ACSL4 contributes to sevoflurane-induced ferroptotic neuronal death in SH-SY5Y cells via the 5′ AMP-activated protein kinase/mammalian target of rapamycin pathway

Lei Cheng et al.

Summary: Downregulation of ACSL4 restrains sev-induced ferroptotic cell death via AMPK/mTOR signaling, providing the basis for an approach to alleviate sev-induced postoperative cognitive dysfunction (POCD). Sev treatment increases Fe2+ levels and decreases mRNA levels of SLC7A11 and GPX4 in SH-SY5Y cells. Silencing ACSL4 could abrogate sev-induced cell damage through increased cell viability and GPX4 protein levels, and decreased iron levels, ROS production, MDA and 4-HNE content.

ATF4 promotes lung cancer cell proliferation and invasion partially through regulating Wnt/β-catenin signaling

Jiang Du et al.

Summary: The study found that ATF4 expression in non-small cell lung cancer is associated with cancer cell growth and invasion, and it participates in cancer progression by promoting the activity of the Wnt signaling pathway. ICG001 can inhibit the function of ATF4, but when ATF4 and ICG001 are combined, the invasion ability of cancer cells significantly increases, indicating that ATF4's function is not solely dependent on the Wnt/beta-catenin signaling pathway.
