Two-Component Redox Organocatalyst for Peptide Bond Formation

Handoko et al.

Summary: Peptides are important therapeutic drugs that can be synthesized automatically. However, the current peptide synthesis methods are not atom economical and require excessive coupling agents and protected amino acids. A recent study proposed a new organic catalyst that can be used in automated peptide synthesis, which reduces the amount of phosphine and eliminates the need for a dehydrating agent by utilizing a new catalytic cycle for amide bond formation.

Radical-Mediated Activation of Esters with a Copper/Selectfluor System: Synthesis of Bulky Amides and Peptides

Akira Matsumoto et al.

Summary: A new approach for ester activation via a radical-mediated process enabled by a copper/Selectfluor system is described here. This method allows for the conversion of para-methoxybenzyl esters into acyl fluorides, which can be directly used in the one-pot synthesis of amides and peptides. The method has been successfully applied to the iterative formation of sterically hindered amide bonds as a proof of concept.

Silicon compounds as stoichiometric coupling reagents for direct amidation

Joshua J. Davies et al.

Summary: Amidation is a common chemical reaction in industry, but there is currently no widely recognized safe, high-yielding, environmentally friendly, and inexpensive amidation protocol. Direct amidation of a carboxylic acid with an amine is seen as a potential solution to this problem. The gradual development and use of silicon reagents for direct amidation since the 1960s is covered in this review, along with the methods published up to April 2021 related to silicon reagent-mediated direct amidation of a carboxylic acid with an amine.

Metal-free approach for hindered amide-bond formation with hypervalent iodine(III) reagents: application to hindered peptide synthesis

Hyo-Jun Lee et al.

Summary: A new bio-inspired approach for amide and peptide synthesis using a-amino esters with a potential activating group (PAG) at the ester residue is reported. The ester functionality is activated under mild metal-free conditions by exploiting the dearomatization of phenols with hypervalent iodine(III) reagents. Highly reactive acyl fluoride intermediates are successfully generated using a pyridine hydrogen fluoride complex, facilitating the smooth formation of sterically hindered amides and peptides from bulky amines and a-amino esters.
