Unmasking the Hidden Carbonyl Group Using Gold(I) Catalysts and Alcohol Dehydrogenases: Design of a Thermodynamically-Driven Cascade toward Optically Active Halohydrins

Sergio Gonzalez-Granda et al.

Summary: A concurrent cascade reaction using a gold(I) N-heterocyclic carbene and an alcohol dehydrogenase was developed for the synthesis of enantiopure halohydrins. The reaction proceeds through regioselective hydration of haloalkynes followed by stereoselective bioreduction of alpha-halomethyl ketone intermediates. The method demonstrates high selectivity and good yields of chloro- and bromohydrins. Additionally, the synthesis of enantiomers of styrene oxide in high yields and optical purities was achieved.

Chemoenzymatic Synthesis of the Most Pleasant Stereoisomer of Jessemal

Silvia Venturi et al.

Summary: This article describes the asymmetric synthesis of the most pleasant enantiomer of Jessemal fragrance, with key steps involving one-pot reduction using stereoselective redox enzymes and regioselective epoxide ring-opening using organocuprate or organolithium nucleophiles.

Chiral Aminoalcohols and Squaric Acid Amides as Ligands for Asymmetric Borane Reduction of Ketones: Insight to In Situ Formed Catalytic System by DOSY and Multinuclear NMR Experiments

Yana Nikolova et al.

Summary: A comparison study between squaric acid amides and chiral aminoalcohols as ligands in a model reduction reaction showed that aminoalcohols exhibited higher efficiency. Mechanistic insights on in situ formation and stability of intermediates generated from ligands and borane were obtained through H-1 DOSY and multinuclear NMR, contributing to a better understanding of the complexity prior to oxazaborolidine formation.

Total Synthesis of Limaol

Stephan N. Hess et al.

Summary: Limaol, a marine-derived C40-polyketide, is unique in structure due to the nonthermodynamic array of four skipped methylene substituents on its hydrophobic tail. This distinctive segment was assembled using a two-directional approach and coupled to the polyether domain through an allyl/alkenyl Stille reaction under neutral conditions. The core region was prepared through several key steps, including asymmetric propargylation catalyzed by 3,3'-dibromo-BINOL, gold-catalyzed spirocyclization, and substrate-controlled allylation to introduce the southern sector.

A cooperative zinc/catalytic indium system for the stereoselective sequential synthesis of (E)-1,3-dienes from carbonyl compounds

Jorge Gonzalez-Rodriguez et al.

Summary: A cooperative zinc/indium system is employed for the stereoselective synthesis of highly functionalized (E)-1,3-dienes through the allylation/elimination reaction of aldehydes and ketones with 1,3-dichloropropene. The methodology offers a broad substrate scope, simple procedure, mild conditions, and ease of operation as its main advantages.
