TIR domains of plant immune receptors are 2′,3′-cAMP/cGMP synthetases mediating cell death

Dongli Yu et al.

Summary: This study reveals that plant Toll/interleukin-1 receptor (TIR) proteins not only have NADase function, but also act as 2',3'-cAMP/cGMP synthetases. These cyclic nucleotide monophosphates (cNMPs) play a critical role in plant immune responses.

TALE-triggered and iTALE-suppressed Xa1-mediated resistance to bacterial blight is independent of rice transcription factor subunits OsTFIIAγ1 or OsTFIIAγ5

Xiameng Xu et al.

Summary: This study found that OsTFIIA gamma 1 and OsTFIIA gamma 5 in rice do not affect TALE-triggered and iTALE-suppressed Xa1-mediated resistance, suggesting the involvement of unknown factors in the interaction of Xa1, TALEs, and iTALEs.

The ZAR1 resistosome is a calcium-permeable channel triggering plant immune signaling

Guozhi Bi et al.

Summary: The study showed that the activated ZAR1 protein can form pentameric complexes in the plasma membrane, exhibiting ion channel activity dependent on the conserved acidic residue Glu11. Pre-assembled ZAR1 resistosome readily incorporated into planar lipid-bilayers and displayed calcium-permeable cation-selective channel activity. Activation of ZAR1 in the plant cell resulted in Glu11-dependent Ca2+ influx, perturbation of subcellular structures, production of reactive oxygen species, and cell death.

Novel assays to monitor gene expression and protein-protein interactions in rice using the bioluminescent protein, NanoLuc

Ken-ichiro Taoka et al.

Summary: Nluc demonstrates higher sensitivity and specificity in rice compared to Fluc. The successful detection of strong response to Hd3a after linking Nluc to OsMADS15 gene suggests its potential application in research involving photoperiodic regulation of flowering.

Identification of TAL and iTAL effectors in Japanese strain T7133 of Xanthomonas oryzae pv. oryzae

Ayaka Yoshihisa et al.

Summary: This study determined the whole genome sequence of Xanthomonas oryzae pv. oryzae (Xoo) strain T7133 and identified TAL and iTAL effectors, showing that the iTAL effector of Xoo T7133 may contribute to overcoming rice Xa1-mediated immunity.

Plant helper immune receptors are Ca2+-permeable nonselective cation channels

Pierre Jacob et al.

Summary: Plant nucleotide-binding leucine-rich repeat receptors (NLRs) play a crucial role in regulating immunity and cell death, with helper NLRs being essential for the function of many sensor NLRs. NRG1.1 and ADR1 are two helper NLRs that mediate cell death through Ca2+ influx, by forming Ca2+-permeable cation channels to directly regulate cytoplasmic Ca2+ levels.
