Neuroinflammation in neurological disorders: pharmacotherapeutic targets from bench to bedside

Awanish Mishra et al.

Summary: Neuroinflammation is a crucial defense mechanism of the nervous system against pathogens and infections, commonly observed in various neurological disorders such as Alzheimer's disease, Parkinson's disease, and Epilepsy. The review highlights recent advancements in understanding the molecular mechanisms of neuroinflammation in these disorders, as well as the current status of anti-inflammatory agents in preclinical and clinical studies. Possible limitations and future directions for the effective use of anti-inflammatory agents in neurological disorders are also discussed.

Selonsertib Alleviates the Progression of Rat Osteoarthritis: An in vitro and in vivo Study

Jiyuan Yan et al.

Summary: The study demonstrated that Selonsertib has anti-arthritic effects in rat OA by preventing inflammatory reactions, cartilage degradation, and cell apoptosis. The protective roles of Ser are mediated through the ASK1/P38/JNK and NFκB pathways. In addition, intra-articular injection of Ser significantly alleviated surgery-induced cartilage damage in the rat OA model.

Development of a Physiologically Based Pharmacokinetic Model of Mitragynine, Psychoactive Alkaloid in Kratom (Mitragyna Speciosa Korth.), In Rats and Humans

Kimheang Ya et al.

Summary: The study developed a PBPK model to predict the distribution of mitragynine in rats and humans, incorporating biological features related to brain, liver, and fat tissue. The model successfully simulated data under different dosing regimens in two species. The developed PBPK models can guide optimal dosing regimens for mitragynine.
