Summary: Bone marrow-mediated trained innate immunity can enhance immune responsiveness, but maladaptive training can lead to inflammatory comorbidities. Periodontitis can induce maladaptive trained phenotype in the bone marrow, resulting in increased production of myeloid cells with inflammatory preparedness. The trained phenotype of the bone marrow can be transmitted to naive recipients through transplantation and lead to increased inflammatory responsiveness and disease severity. Inhibiting IL-1 signaling can prevent maladaptive training caused by periodontitis.

ACTIVATE-2: A Double-Blind Randomized Trial of BCG Vaccination Against COVID-19 in Individuals at Risk

Summary: A recent study shows that Bacillus Calmete-Guerin (BCG) vaccination reduces the occurrence of new infections, particularly respiratory infections, in the elderly. Another study confirms the efficacy of BCG vaccination against COVID-19, with a 68% reduction in the risk of developing the disease compared to placebo. BCG vaccination also leads to a higher rate of positive anti-SARS-CoV-2 antibodies.

Vaccination in patients with kidney failure: lessons from COVID-19

Summary: Infection is a major cause of death in patients with chronic kidney disease (CKD). However, vaccination efficacy is reduced in CKD patients due to immune alterations caused by kidney dysfunction and chronic inflammation. COVID-19 has detrimental effects in CKD patients, making successful vaccination against it crucial. While some CKD patients may not generate humoral immunity after vaccination, their vaccine-specific T cell response may still provide protection against severe COVID-19. Establishing protection correlates for CKD patients is urgently needed for personalized vaccination strategies.

Assessing the Functional Heterogeneity of Monocytes in Human Septic Shock: a Proof-of-Concept Microfluidic Assay of TNFα Secretion

Summary: The study evaluated the behavior of circulating monocytes from septic shock patients using an advanced microfluidic assay combined with fluorescence microscopy. Results showed that TNF alpha secretion by single monocytes was significantly decreased in septic patients, with a dampened production shortly after LPS stimulation. The microfluidic assay revealed heterogeneous individual behavior of monocytes from septic shock patients, while both healthy donors and patients exhibited similar phagocytic activities over time.
