Genetic screening for single-cell variability modulators driving therapy resistance

Eduardo A. Torre et al.

Summary: A genetic screen identified a new mode of altering resistance to BRAF inhibition in melanoma by increasing cellular plasticity and pushing cells towards a more differentiated state. Modulating cellular plasticity may alter cell fate decisions and prove useful for treating drug resistance in other cancers.

Cellpose: a generalist algorithm for cellular segmentation

Carsen Stringer et al.

Summary: Cellpose is a generalist, deep learning-based segmentation method that can precisely segment cells from various types of images without requiring retraining or parameter adjustments. It was trained on a highly diverse image dataset and supports a three-dimensional extension. Software has been developed to support community contributions to the training data.

Variability within rare cell states enables multiple paths toward drug resistance

Benjamin L. Emert et al.

Summary: Differences in molecular features among individual cells can lead to dramatic differences in cell fate, such as the survival or death of cancer cells upon drug treatment. The Rewind methodology combines genetic barcoding with RNA fluorescence in situ hybridization to capture rare cells that give rise to cellular behaviors of interest. By applying Rewind to BRAF(V600E) melanoma, researchers traced drug-resistant cell fates back to single-cell gene expression differences in their drug-naive precursors, revealing hidden, rare-cell variability that underlies a range of latent phenotypic outcomes upon drug exposure.
