Signaling metabolite L-2-hydroxyglutarate activates the transcription factor HIF-1α in lipopolysaccharide-activated macrophages

Niamh C. Williams et al.

Summary: Activated macrophages undergo metabolic reprogramming, in which the metabolites such as 2HG play a role in regulating their function. In LPS-activated macrophages, both enantiomers of 2HG, L-2HG and D-2HG, are accumulated. L-2HG can promote the expression of IL-1 beta and induce an inflammatory metabolic state. These changes may be mediated through activation of HIF-1 alpha by L-2HG, and decreased expression of L-2HGDH may contribute to L-2HG accumulation.

A matter of time: temporal structure and functional relevance of macrophage metabolic rewiring

Gretchen L. Seim et al.

Summary: The response of macrophages to stimulation involves dynamic rewiring of metabolism to support important cellular processes during immune response. Recent studies have shown that this metabolic rewiring is temporally structured. We review the regulatory mechanisms and functional implications of these metabolic dynamics.

D-2-Hydroxyglutarate is an anti-inflammatory immunometabolite that accumulates in macrophages after TLR4 activation

Kyra E. de Goede et al.

Summary: Upon activation, macrophages undergo metabolic rewiring and accumulate immunometabolite 2-hydroxyglutarate (2HG), which plays a regulatory role in macrophage function and can inhibit inflammatory responses. This study reveals a negative feedback mechanism involving the immunomodulatory metabolite D-2HG, which may exert distinct anti-inflammatory effects.

Itaconate confers tolerance to late NLRP3 inflammasome activation

Monika Bambouskova et al.

Summary: Itaconate, induced upon TLR stimulation, regulates NLRP3 inflammasome activation and synergizes with iNOS, establishing tolerance to NLRP3 inflammasome in inflammatory macrophages. Mechanistically, itaconate prevents full caspase-1 activation and gasdermin D processing, potentially leading to pyroptotic cell death and tissue damage if not controlled.

Control of endothelial quiescence by FOXO-regulated metabolites

Jorge Andrade et al.

Summary: Andrade et al. demonstrate that FOXO1 regulates mitochondrial metabolism to produce the metabolite S-2HG, promoting a quiescent endothelial state. S-2HG inhibits cell cycle progression and vascular expansion, highlighting its role as a key metabolite in regulating endothelial quiescence.
