Influenza Polymerase Inhibitors: Mechanisms of Action and Resistance

Emi Takashita

Summary: The RNA-dependent RNA polymerase of influenza virus is highly conserved among different types of influenza viruses and is considered a key target for antiviral agents. Several polymerase inhibitors targeting different subunits have been developed, including PB1 inhibitor favipiravir, PB2 inhibitor pimodivir, and PA inhibitor baloxavir marboxil, which have shown antiviral activity and are in late-phase clinical trials for influenza treatment.

REGN-EB3: First Approval

Anthony Markham

Summary: REGN-EB3, a combination of three fully-human monoclonal antibodies targeting Ebola virus glycoprotein, was recently approved by the US FDA based on the PALM study conducted during an Ebola outbreak in the Democratic Republic of Congo. This marks a significant milestone in the development of REGN-EB3 as a treatment for Zaire ebolavirus infection in adult and paediatric patients.

Impacts of the Filoviridae family

Sylvester Languon et al.

Summary: Since the discovery of the first member of the Filoviridae family, there have been over 40 reported outbreaks of filovirus diseases, with most of them occurring in Africa. These outbreaks not only result in deaths (primary impacts), but also have secondary impacts on health, social, economic, and political aspects. The recent large outbreaks in West and Central Africa highlight the urgent need to strengthen the capacity of health services in disease hotspots.

Structural basis for inhibition of the SARS-CoV-2 RNA polymerase by suramin

Wanchao Yin et al.

Summary: Suramin, an old drug, serves as a potent inhibitor of the SARS-CoV-2 RNA-dependent RNA polymerase by blocking RNA binding. It is at least 20 times more effective than the approved nucleotide drug for COVID-19 treatment, remdesivir. The cryo-electron microscopy structure of the viral RdRp complexed with suramin reveals its mechanism of action in inhibiting viral replication.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Resurgence of Ebola virus in 2021 in Guinea suggests a new paradigm for outbreaks

Alpha Kabinet Keita et al.

Summary: The 2021 Ebola virus outbreak in Guinea was found to be a resurgence from the previous outbreak, rather than a new spillover event from an animal reservoir. This highlights the need for long-term medical and social care for survivors to reduce the risk of re-emergence and prevent further stigmatization.
