Combination strategies with PD-1/PD-L1 blockade: current advances and future directions

Ming Yi et al.

Summary: Antibodies targeting PD-1/PD-L1 revive immune response against cancer cells, but the low response rate calls for combination therapies. Combining with chemotherapy, radiotherapy, and other treatments can achieve better outcomes, and individualized combination selection is needed to overcome treatment resistance.

Combining mannose receptor mediated nanovaccines and gene regulated PD-L1 blockade for boosting cancer immunotherapy

Jie Chen et al.

Summary: This study explores an intensive tumor immunotherapeutic strategy by combining mannosylated nanovaccines with gene regulated PD-L1 blockade. The mannose modified nanovaccines can effectively co-encapsulate antigen and adjuvant, promoting endocytosis and cross presentation in dendritic cells. However, the limited inhibition of tumor growth by nanovaccines in the immunosuppressed tumor microenvironment can be overcome by combining with gene regulated PD-L1 blockade, leading to significant tumor remission.

In situ phase transitional polymeric vaccines for improved immunotherapy

Jie Wang et al.

Summary: A new strategy is proposed in this study to improve the delivery of cancer vaccines by dynamic size modulation, enhancing lymph node drainage and dendritic cell uptake. The results showed that this strategy induced a rapid and robust immune response in a mouse model, with a stronger humoral immune response observed when immunized with MHC-II restricted antigen.

Th17 cells contribute to combination MEK inhibitor and anti-PD-L1 therapy resistance in KRAS/p53 mutant lung cancers

David H. Peng et al.

Summary: Understanding the resistance mechanisms in mutant KRAS lung cancers to targeted therapies and immune checkpoint blockade is crucial for developing novel combination therapies. MEK inhibition combined with anti-PD-L1 can synergistically reduce lung tumor growth, but resistance may develop eventually. Increased Th17 cell infiltration in resistant tumors promotes invasiveness and resistance to MEK inhibitors, and a triple therapy targeting MEK, PD-L1, and IL-17 may overcome this resistance for better in vivo responses.

Photothermal-Chemotherapy Enhancing Tumor Immunotherapy by Multifunctional Metal-Organic Framework Based Drug Delivery System

Weidong Ni et al.

Summary: Immunotherapy shows promise for tumor treatment, but its clinical effect is limited by tumor immunogenicity and the immunosuppressive microenvironment. Researchers combined metal-organic framework MIL-100 loaded with mitoxantrone (MTO) with photothermal-chemotherapy to enhance immunogenic cell death. The resulting MMH NPs, with dual therapeutic properties and imaging modes, showed high antitumor efficacy and immune effect when co-injected with anti-OX40 antibody in colorectal cancer. This novel combined therapeutic strategy is promising for future tumor therapy.

Targeting dual gene delivery nanoparticles overcomes immune checkpoint blockade induced adaptive resistance and regulates tumor microenvironment for improved tumor immunotherapy

Yuanji Feng et al.

Summary: This study developed a revolutionary dual gene delivery system loading pshVEGF-A and pshPD-L1 to overcome adaptive resistance to ICB therapy, achieving effective anti-tumor immunotherapy. The strategy eliminates adaptive resistance, achieves tumor vessel normalization, and reprograms the tumor immune microenvironment, contributing to the diversity of immune combination therapy.

Combination of epigenetic regulation with gene therapy-mediated immune checkpoint blockade induces anti-tumour effects and immune response in vivo

Huapan Fang et al.

Summary: While immunotherapy has shown promise as a cancer treatment, durable benefits are often limited by immune escape mechanisms. This study combines epigenetic regulation with gene therapy for immune checkpoint blockade to enhance anti-tumor effects and immune responses in vivo, providing a potential strategy for tumor treatment and prevention of relapse and metastasis.
