Dietary Flavonoids Alleviate Inflammation and Vascular Endothelial Barrier Dysfunction Induced by Advanced Glycation End Products In Vitro

Summary: This study aimed to compare the protective effects of three dietary flavonoids (A7G, I3R, and C3G) on AGEs-induced inflammation and vascular endothelial dysfunction, and analyzed the potential mechanisms using molecular docking analysis. Results showed that C3G had the best inhibition on inflammatory cytokines, while I3R had the best effect in improving endothelial dysfunction. This study provides knowledge and suggestions regarding the benefits of dietary flavonoids in diabetic vascular complications.

Identification of novel potential PI3Kα inhibitors for cancer therapy

Summary: This study identified a novel potential PI3Kα inhibitor, TCM-N1, through structure-based virtual screening, bioassays, and molecular dynamics simulations. TCM-N1 exhibited superior binding affinity and moderate inhibition activity against PI3Kα, as well as relatively high anti-tumor proliferation ability in gastric, colorectal, and cervical cancer cells. The compound was tightly embedded into the ATP-binding pocket via hydrogen bonds, van der Waals interactions, and hydrophobic interactions, suggesting promise for the development of more potent PI3Kα-inhibiting analogs.

Protective effects of three structurally similar polyphenolic compounds against oxidative damage and their binding properties to human serum albumin

Summary: Brazilin, hematoxylin, and hematein are polyphenols with similar structures and rich biological activities, and they have protective effects against oxidative degradation of human serum albumin. The biological effects of these polyphenols are mainly influenced by their binding to protein, and fluorescence experiments indicate that they bind to protein through a static quenching mechanism, causing some conformational changes. These findings provide insights into the role of molecular structure in the protective effects and binding abilities of polyphenols against protein oxidative damage.
