Preserving mitochondrial function by inhibiting GRP75 ameliorates neuron injury under ischemic stroke

Bin Wen et al.

Summary: Ischemic stroke is closely related to mitochondrial dysfunction, with mitochondrial calcium overload playing a key role in the pathophysiological process. Inhibiting GRP75 can effectively ameliorate mitochondrial function and potentially provide evidence for translational studies of ischemic diseases.

Neuronal Death Mechanisms and Therapeutic Strategy in Ischemic Stroke

Rui Mao et al.

Summary: The mechanisms of neuronal death following ischemic stroke involve various pathological events, including excitotoxicity, oxidative stress, and inflammatory responses. Understanding the mechanisms of these events is critical for developing new neuroprotective strategies, and some progress has been made in pharmacological research.

Oxidative Stress in Ischemia/Reperfusion Injuries following Acute Ischemic Stroke

Anamaria Jurcau et al.

Summary: This article discusses the application of recanalization therapy in the treatment of acute ischemic stroke and the potential ischemia/reperfusion injuries it may cause. Oxidative stress and mitochondrial dysfunction have been identified as significant factors in the pathogenesis of these injuries, but clinical studies targeting oxidative pathways have mostly been unsuccessful.

Non-mitogenic fibroblast growth factor 1 protects against ischemic stroke by regulating microglia/macrophage polarization through Nrf2 and NF-ΚB pathways

Confidence Dordoe et al.

Summary: This study evaluated the neuroprotective effects of nmFGF1 on microglia in a mouse stroke model and determined its mechanism of action. The findings showed that nmFGF1 reduced infarct size and improved neurological deficits by regulating the secretion of inflammatory and anti-inflammatory cytokines. In vitro experiments demonstrated that nmFGF1 modulated the expression of these cytokines under oxygen-glucose deprivation and lipopolysaccharide stimulation. Furthermore, nmFGF1 activated the Nrf2 pathway and inhibited the NF-kappa B pathway, leading to a reduction in neuroinflammation. These results suggest that nmFGF1 has the potential to be used as a therapeutic agent for ischemic stroke.

Summary: Intracranial hemorrhage (ICH) is a devastating disease categorized into primary brain injury (PBI) and secondary brain injury (SBI), with oxidative and inflammatory stresses playing central roles. Nrf2 activators have the therapeutic potential in treating ICH patients by up-regulating anti-oxidative enzymes in the brain.

In vivo dynamics of acidosis and oxidative stress in the acute phase of an ischemic stroke in a rodent model

Ilya Kelmanson et al.

Summary: Ischemic cerebral stroke is a major cause of death and disability in humans, but the molecular processes underlying its development are poorly understood. The study recorded the dynamics of intracellular pH and H2O2 during I/R and found a significant difference in the metabolic processes between in vivo and cultured neuron models. The findings highlight the limitations of cell culture as a predictor of metabolic events in vivo.

The Nrf2 Pathway in Ischemic Stroke: A Review

Marcelo Farina et al.

Summary: Ischemic stroke, the sudden loss of blood flow in specific area(s) of the brain, is a leading cause of permanent disability and death worldwide. The only approved pharmacological treatment, intravenous thrombolysis with recombinant tissue plasminogen activator, has limitations and does not address the complex oxidative events in the ischemic cascade. The transcription factor Nrf2 has been identified as a potential target to mitigate oxidative events and research has shown its protective role in experimental models of ischemic stroke.

Summary: Stroke is the second most common cause of death globally and the leading cause of death in China. Oxidative stress plays a crucial role in the pathogenesis of cerebral ischemia injury, making antioxidants a potential therapeutic target for ischemic stroke. Some polyphenols have been found to have strong antioxidant effects on reactive oxygen species generation and oxidative stress after ischemic stroke.
