Targeted Alpha Therapy of Glioma Using 211At-Labeled Heterodimeric Peptide Targeting Both VEGFR and Integrins

In vitro and in vivo evaluation of 211At-labeled fibroblast activation protein inhibitor for glioma treatment

Summary: This study investigated the therapeutic effect of At-211-labeled fibroblast activation protein inhibitor (FAPI) for glioma treatment. The results showed that At-211-FAPI-04 had fast and specific binding to FAP-positive U87MG cells and significantly reduced cell viability, arrested cell cycle, and suppressed cell proliferative efficacy. Intratumoral injection of At-211-FAPI-04 resulted in 6-fold higher accumulation in tumor sites compared to intravenous injection. At-211-FAPI-04 effectively suppressed tumor growth and prolonged median survival in U87MG xenografts without obvious toxicity to normal organs. These findings suggest that targeted alpha-particle therapy mediated by At-211-FAPI-04 is a promising strategy for glioma treatment.
