Discovery of a novel DDRs kinase inhibitor XBLJ-13 for the treatment of idiopathic pulmonary fibrosis

Summary: Idiopathic pulmonary fibrosis (IPF) is a chronic and fatal lung disease characterized by destruction of lung tissue and extracellular matrix deposition. Known drugs for IPF are insufficient, and there is an urgent need for validation of new drug targets. Tyrosine kinase receptor DDRs has been identified as a potential therapeutic target for pulmonary fibrosis. In this study, a new indazole derivative XBLJ-13 was synthesized and shown to be a highly specific and potent DDRs inhibitor with anti-inflammatory and anti-fibrotic activities. Extensive profiling revealed that XBLJ-13 had low toxicity and better pharmacokinetic properties compared to nintedanib, and showed promising efficacy in treating pulmonary fibrosis in mice.

A novel DDR1 inhibitor compound 2 was designed using a scaffold-based molecular design workflow, and it showed excellent selectivity and inhibition of DDR1 as well as promising therapeutic effects in a mouse colitis model.

Design of Rational JAK3 Inhibitors Based on the Parent Core Structure of 1,7-Dihydro-Dipyrrolo [2,3-b:3′,2′-e] Pyridine

Summary: JAK3 is a promising target for autoimmune disease treatment, but its selective targeting is challenging due to high homology with other JAK family members. This study designed a series of 1,7-dihydro-dipyrrolo [2,3-b:3 ',2 '-e] pyridine derivatives using the backbone growth method and identified 14 JAK3 inhibitors. Molecular dynamics simulations confirmed the structural stability of these inhibitors. Analysis showed that binding efficiency was achieved mainly through electrostatic and VDW interactions.
