2-substituted tricyclic oxazolo[5,4-d]pyrimidine library: Design, synthesis, and cytotoxicity activity

Synthesis and Anticancer Activity of Podophyllotoxin Derivatives

Summary: Two series of podophyllotoxin derivatives were synthesized by adding a 4 beta-sulfanilamide or substituting a 4 beta-amide into podophyllotoxin. The derivative 9f (2-thienoylaminoepipodophyllotoxin) showed the highest cytotoxic activity against cancer cells, especially inhibiting the growth of MCF-7 cells. Modulation of the morphology of MCF-7 cells treated with 9f further confirmed its cytotoxic activity against cancer cells.

Summary: An efficient protocol for synthesizing fused quinazolinones through oxidative free-radical cyclization under metal- and tin-free conditions is described. The study focuses on the oxidative cyclization of various N-3-omega-iodoalkyl derivatives to provide tricyclic systems using dicumyl peroxide as the sole reagent. This method has been successfully applied to the synthesis of various fused quinazolinone analogues, including natural products, and new conjugates as well as the total synthesis of leucomidine C were reported.

Summary: At 100 K, the compound crystallizes in the orthorhombic space group Pna21 with two similar molecules in the asymmetric unit. Intermolecular pi-pi stacking and C=O···π interactions link molecules into stacks parallel to [100]. The crystal packing is mainly contributed by H···H (49.4%) and H···O/O···H (21.5%) interactions, with a significant dispersion energy contribution. The crystal was refined as a two-component inversion twin.
