Fatty Acids, CD36, Thrombospondin-1, and CD47 in Glioblastoma: Together and/or Separately?

Summary: Glioblastoma (GBM) is an aggressive tumor with high heterogeneity. The accumulation of fatty acid metabolites is associated with low survival rates in patients with high-grade gliomas. The diversity of brain lipids, especially polyunsaturated fatty acids (PUFAs), and the involvement of FA transport proteins and FA translocases are crucial for PUFAs transportation through the blood-brain barrier and delivery to brain cells. Among them, the CD36 FA translocase plays a role in the uptake of long-chain FAs and oxidized lipoproteins. Furthermore, the interaction between CD36 and thrombospondin-1 (TSP-1) has an important role in glioma cell invasion and angiogenesis in GBM. The interconnection and interrelationship between FA, CD36, CD47, and their ligand TSP-1 have been highlighted in recent research to exert a cumulative effect on the modulation of the GBM molecular network.

Targeting the gp130/STAT3 Axis Attenuates Tumor Microenvironment Mediated Chemoresistance in Group 3 Medulloblastoma Cells

Summary: Microglia play a crucial role in promoting acquired drug resistance in Group 3 medulloblastoma (MB) by secreting IL-6, which activates the JAK1/STAT3 pathway and enhances chemoresistance. The IL-6 family cytokines, including IL-6, OSM, LIF, and IL-11, contribute to the feedback network of cytokine secretion and further enhance JAK1/STAT3 activity and chemoresistance in MB cells. Targeting the gp130/JAK/STAT pathway can effectively overcome treatment resistance in Group 3 MB.

Single-cell landscapes of primary glioblastomas and matched explants and cell lines show variable retention of inter- and intratumor heterogeneity

Summary: Glioblastomas (GBMs) are highly heterogeneous brain tumors, and patient-derived models have been used to study this heterogeneity. However, the extent to which these models recapitulate GBM genomic features is still unclear. In this study, the researchers analyzed the genomes and transcriptomes of GBM tumors and patient-derived models, and found that the models exhibit genetic similarities to the tumors and retain varying gene expression characteristics. These findings provide valuable insights for using GBM-derived models to study cellular heterogeneity.

Palmitoyl transferases act as potential regulators of tumor-infiltrating immune cells and glioma progression

Summary: In this study, the inhibition of ZDHHCs was found to suppress glioma-cell viability and autophagy, promote apoptosis, and increase the sensitivity of glioma cells to chemotherapy. Additionally, targeting ZDHHCs weakened the migratory ability of microglia induced by glioma cells. These findings suggest that targeting ZDHHCs may be a promising approach for glioma treatments.

Hypomethylation-induced prognostic marker zinc finger DHHC-type palmitoyltransferase 12 contributes to glioblastoma progression

Summary: This study conducted expression and prognostic analyses of the zinc finger DHHC-type palmitoyltransferase (ZDHHC) family in gliomas. ZDHHC12 was identified as the only unfavorable prognostic marker and its knockdown significantly inhibited the growth, migration, and invasion capabilities of glioma cells. Further molecular mechanism analysis revealed that decreased DNA methylation may contribute to the upregulation of ZDHHC12 expression. This study contributes to a deeper understanding of glioma progression and therapeutics.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
