Detailed Molecular Mechanisms Involved in Drug-Induced Non-Alcoholic Fatty Liver Disease and Non-Alcoholic Steatohepatitis: An Update

Laura Giuseppina Di Pasqua et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis (NASH) are global public health challenges. Drug-induced hepatic steatosis (DIHS) is a major cause of NAFLD/NASH, primarily through interfering with mitochondrial activity and inhibiting fatty acid oxidation.

Chronic AT1 blockade improves hyperglycemia by decreasing adipocyte inflammation and decreasing hepatic PCK1 and G6PC1 expression in obese rats

Ruben Rodriguez et al.

Summary: The study demonstrates that chronic AT(1) blockade improves obesity-related hyperglycemia in rats by enhancing adipocyte function and reducing hepatic glucose production via gluconeogenesis. The results shed light on the therapeutic potential of chronic AT(1) blockade for hyperglycemia.

Update on New Aspects of the Renin-Angiotensin System in Hepatic Fibrosis and Portal Hypertension: Implications for Novel Therapeutic Options

Indu G. Rajapaksha et al.

Summary: The renin angiotensin system (RAS) plays a crucial role in liver fibrosis and portal hypertension, with the classical RAS promoting fibrosis while the alternate RAS counteracts its harmful effects. Overexpression of ACE2 can reduce liver fibrosis, and blockers targeting alternate RAS receptors could be useful in treating portal hypertension.

Increased Hepatic Lipogenesis Elevates Liver Cholesterol Content

Jean-Mathieu Berger et al.

Summary: This study found that hepatic lipogenesis induced by FF/HS diet increased liver triglyceride and cholesterol contents in mice. The increased cholesterol content in the liver was mainly due to reduced excretion of bile acids, reduced fecal cholesterol excretion, and increased cholesterol uptake from plasma lipoproteins. Additionally, the study showed that dietary cholesterol intake and cholesterol synthesis rates alone cannot explain the hypercholesterolemia associated with NAFLD, and controlling fatty acid synthesis may be important for managing dyslipidemia.
