The Acid-Base/Deprotonation Equilibrium Can Be Studied with a MicroScale Thermophoresis (MST)

Pawel Mateusz Nowak et al.

Summary: This study is a proof-of-concept of using MicroScale thermophoresis (MST) for the analysis of acid-base dissociation and pK(a) constant determination. The results show that the obtained pK(a) values are consistent with reference methods, indicating the analytical potential and utility of MST technology. Future development of a new detection system for thermophoretic movement could further enhance its applications.

Extension of the Internal Standard Method for Determination of Thermodynamic Acidity Constants of Compounds Sparingly Soluble in Water by Capillary Zone Electrophoresis

Lucie Nytrova et al.

Summary: This study extends the applicability of the internal standard method published in 2009, demonstrating that the original capillary zone electrophoresis method can effectively overcome analyte interactions in mixed solvents. The extended method is illustrated on six selected beta-blockers.

Microscale thermophoresis for studying protein-small molecule affinity: Application to hyaluronidase

Rouba Nasreddine et al.

Summary: Microscale thermophoresis (MST) is a sensitive biophysical tool that allows rapid and accurate determination of the dissociation constant (K-d) between enzymes and small molecules, aiding in the study of binding affinity between biomolecules. The study demonstrated that hyaluronidase maintained enzymatic activity after fluorescent labelling, but showed no affinity with flavonoid compounds under the tested conditions.

Kinetic Microscale Thermophoresis for Simultaneous Measurement of Binding Affinity and Kinetics

Julian A. C. Stein et al.

Summary: MST is a versatile technique for measuring binding affinities, based on directional movement of molecules in a temperature gradient. We extended MST to measure binding kinetics by increasing thermal dissipation, allowing for simultaneous determination of kinetics and binding affinity. By measuring DNA hybridization rates, we observed linear dependence of on-rates on salt concentration and weak dependence on strand length and temperature.

Microscale Thermophoresis and additional effects measured in NanoTemper Monolith instruments

Blanca Lopez-Mendez et al.

Summary: NanoTemper Monolith instruments are widely used in academia and industry for measuring molecular interactions. The technology has been extensively reviewed, and some assumptions and limitations have been revised in the last decade. This letter presents the experience gathered in scientific service facilities and highlights possible pitfalls for scientists to promote knowledge and good practice in the scientific community.
