Summary: Upstream process development focuses on optimizing media formulations to enhance cell culture conditions and regulate product quality attributes. Transition metal ions in cell culture media, such as Mn, Fe, Cu, and Zn, have significant impacts on cell growth, metabolism, and product quality, but their associations and effects are not well characterized. Equilibrium models can be used to explore metal speciation, but experimental determination of equilibrium constants and consideration of reaction kinetics are also necessary to fully understand the complex interactions between media components and bioprocess performance.

Effect of vitamins and metal ions on productivity and charge heterogeneity of IgG1 expressed in CHO cells

Summary: Recombinant monoclonal antibodies expressed in CHO cells undergo important post-translational modifications which impact product quality. This study investigates the effects of vitamins and metal ions on protein expression and charge heterogeneity. The results show that different components have varied effects on productivity and charge variants, providing information that could be useful for process control and ensuring consistent quality in biosimilar production.

By incorporating in-line monitoring, data analytics, and mechanistic models, bioprocess DT of cell cultures can virtually mirror behaviors under adjustable process conditions. This review highlights recent advances in bioprocess DT components and discusses current challenges and future research directions.
