Stabilization of Gold Nanoparticles by Hairpin DNA and Implications for Label-Free Colorimetric Biosensors

Summary: Gold nanoparticles (AuNPs) have unique optical properties and are widely used in biosensors. This study investigated the effect of hairpin DNA structure and sequence on the stabilization of AuNPs and its implications in AuNP-based label-free colorimetric biosensors. The results showed that hairpin DNA with a tail provided optimal protection for AuNPs, and the flexible tail region favored adsorption to AuNPs. The sensitivity of a DNA sensing assay was doubled when using hairpin DNA with a tail compared to tail-free hairpin.

Summary: A facile method for preparing a gold nanostar-graphene quantum dot composite was reported, which resulted in highly active surfactant-free gold nanostars. The composite was used in a new sensing strategy for colorimetric detection of cysteine by studying its etching reaction with Na2SO3. Larger gold nanostars showed greater activity in the etching reaction, and pre-added cysteine protected the nanostars from etching, allowing for sensitive detection of cysteine with a detection limit of 0.35 nM.
