Activating α7nAChR ameliorates abdominal aortic aneurysm through inhibiting pyroptosis mediated by NLRP3 inflammasome

Hui Fu et al.

Summary: This study found that activating alpha 7 nicotinic acetylcholine receptor (alpha 7nAChR) can slow down the formation of abdominal aortic aneurysm (AAA) by inhibiting pyroptosis mediated by NLRP3 inflammasome, suggesting that alpha 7nAChR could potentially be a pharmacological target for treating AAA.

Role of Oxidative Stress in the Pathogenesis of Atherothrombotic Diseases

Giovanna Petrucci et al.

Summary: Oxidative stress caused by the imbalance between reactive oxygen species formation and antioxidant scavenger system's activity plays a significant role in the development and complications of atherosclerotic cardiovascular diseases (ASCVD). Overexpression of ROS-generating enzymes and uncontrolled ROS formation are associated with accelerated atherosclerosis in animal models, while overexpression of ROS scavenger systems reduces or stabilizes atherosclerotic lesions. Circulating biomarkers derived from lipid and protein oxidation have been identified as independent predictors of ASCVD in humans. Antioxidant supplementation in animal models reduces ROS and attenuates atherosclerotic lesions, but large human trials or prospective studies have failed to show athero-protective effects of antioxidant supplementation. However, long-term consumption of antioxidant-rich diets has been shown to reduce ASCVD in humans.

Pleiotropic effects of alpha-ketoglutarate as a potential anti-ageing agent

Maria M. Bayliak et al.

Summary: AKG, an intermediate of the tricarboxylic acid cycle, plays diverse roles in cellular metabolism and regulation, with potential anti-aging effects demonstrated in experimental models. Its geroprotective actions may involve epigenetic regulation, antioxidant properties, and mimicking caloric restriction and hormesis. AKG's influence on mitochondrial respiration and induction of ROS production could lead to beneficial biological effects according to hormesis hypothesis, despite discrepancies in different models and limitations as a geroprotective drug.

Lithium chloride represses abdominal aortic aneurysm via regulating GSK3?/SIRT1/NF-?B signaling pathway

Tong Xu et al.

Summary: The study found that lithium chloride prevented the development of AAA by inhibiting inflammation, MMPs, and superoxide production, as well as facilitating elastin synthesis. The beneficial effects of lithium chloride may be mediated through the regulation of the GSK3?/SIRT1/NF-?B cascade.

Alpha-ketoglutarate ameliorates pressure overload-induced chronic cardiac dysfunction in mice

Dongqi An et al.

Summary: Alpha-ketoglutarate (AKG) shows potential as a therapeutic agent for heart failure by reducing myocardial hypertrophy and fibrosis, improving cardiac systolic dysfunction, and promoting mitophagy to clear damaged mitochondria and reduce ROS production. Studies suggest that AKG could enhance heart function and delay the progression of heart failure via these mechanisms.

Interplay Between Reactive Oxygen/Reactive Nitrogen Species and Metabolism in Vascular Biology and Disease

Masuko Ushio-Fukai et al.

Summary: Reactive oxygen species (ROS) and reactive nitrogen species (RNS) play important roles as signaling molecules at physiological levels, but excess levels are associated with cardiovascular disease. Cells regulate ROS and RNS levels by controlling metabolic pathways, with implications for disease treatment strategies.
