Nanocatalyst-Mediated Chemodynamic Tumor Therapy

Lu Zhang et al.

Summary: Chemodynamic therapy (CDT) is a novel cancer therapeutic strategy using a Fenton or Fenton-like reaction to generate highly cytotoxic hydroxyl radicals (·OH) from endogenous hydrogen peroxide (H2O2) to kill cancer cells, but its low catalytic efficiency and off-target side effects limit its biomedical application, so various strategies are needed to enhance its therapeutic effect.

Ferrocene-containing polymersome nanoreactors for synergistically amplified tumor-specific chemodynamic therapy

Yuheng Wang et al.

Summary: By loading GOD and TPZ in responsive polymersome nanoreactors, tumor acidic microenvironment can trigger the release of TPZ prodrugs, leading to the conversion of oxygen and glucose into H2O2 under the catalysis of ferrocene and subsequent generation of (OH)• radicals. This synergistic effect efficiently kills tumor cells and suppresses tumor growth, demonstrating the potential of ferrocene-containing responsive polymersome nanoreactors as a potent nano-platform for amplified CDT.

Sustainable amorphous Fenton nanosystem for visualization-guided synergistic tumor elimination

Jingjing Wang et al.

Summary: CDT using SAFN can sustainably produce center dot OH in tumor microenvironment, achieving enhanced tumor therapy. The cascade catalysis and sustained supply of Fe(II) and H2O2 in SAFN contribute to long-lasting center dot OH production for synergistic tumor treatment. The synergistic effect of starvation/LT-PTT/ECDT strategy in SAFN demonstrates remarkable antitumor efficacy.

Renal Clearable Ultrasmall Single-Crystal Fe Nanoparticles for Highly Selective and Effective Ferroptosis Therapy and Immunotherapy

Huan Liang et al.

Summary: This study synthesized ultrasmall single-crystal Fe nanoparticles that showed high activity in inducing tumor cell ferroptosis and immunogenetic cell death in the tumor microenvironment at a low concentration. These nanoparticles effectively suppressed tumor growth, promoted dendritic cell maturation, and triggered T cell response in animal models, and when combined with immune checkpoint blockade therapy, they greatly enhanced the immune response and developed strong immune memory. Furthermore, the nanoparticles were quickly excreted in the kidneys without side effects in normal tissues.
