Nucleosides and emerging viruses: A new story

Vincent Roy et al.

Summary: Nucleoside and nucleotide analogs are important drugs for antiviral treatments, but there are currently no available vaccines or small antivirals for (re)emerging RNA viruses. Thus, the recently developed ribonucleoside analogs have the potential to lead to the discovery of new antiviral nucleosides.

Site-Selective Late-Stage C-H Functionalization via Thianthrenium Salts

Florian Berger et al.

Summary: C-H functionalization is an efficient method to increase the complexity of organic molecules rapidly, but improving selectivity remains a challenge. By utilizing sulfonium salts, we have developed a highly selective approach for aromatic C-H functionalization, enhancing not only the efficiency of the reaction but also expanding the scope of follow-up reactions.

Recent Advances in Thianthrenation/Phenoxathiination Enabled Site-Selective Functionalization of Arenes

Xiao-Yue Chen et al.

Summary: Selective functionalization of simple arenes has been a major challenge due to the similarity of multiple C-H bonds in the same molecule. However, recent research has shown that the use of aryl thianthrenium salts can achieve site-selective C-H functionalization in arenes, providing a promising solution for late-stage functionalization of bioactive molecules.

Environmentally sensitive fluorescent nucleoside analogues as probes for nucleic acid - protein interactions: molecular design and biosensing applications

Dmytro Dziuba

Summary: Fluorescent nucleoside analogues (FNAs) are important tools in studying the interactions of nucleic acids with nucleic acid-binding proteins. They enable real-time monitoring of binding interactions in solutions, with high sensitivity. FNAs are also used to study the conformational dynamics of biomolecular complexes. Therefore, FNAs have potential applications in fundamental biological research and molecular diagnostics drug discovery.

Nickel-Catalyzed Negishi-Type Arylation of Trialkylsulfonium Salts

Hiroko Minami et al.

Summary: The Negishi-type arylation of trialkylsulfonium salts with arylzinc reagents has been achieved under nickel catalysis. The use of cyclohexanethiol as an additional ligand was found to be crucial for promoting C-S cleavage. Mechanistic experiments suggest that the reaction proceeds through single-electron transfer to generate a stable carbon-centered radical, with the thiolate ligand facilitating C-S cleavage and radical recombination.

Antiviral Activity of 7-Substituted 7-Deazapurine Ribonucleosides, Monophosphate Prodrugs, and Triphoshates against Emerging RNA Viruses

Nemanja Milisavljevic et al.

Summary: A series of 7-deazaadenine ribonucleosides and their derivatives were synthesized and tested for antiviral activity against various RNA viruses. The modified triphosphates showed good antiviral activity by inhibiting viral RNA-dependent RNA polymerases, while nucleosides with bulkier heterocycles had lower toxicity.

Late-Stage Heteroarylation of Hetero(aryl)sulfonium Salts Activated by α-Amino Alkyl Radicals

Eva Maria Alvarez et al.

Summary: This study presents a novel method for late-stage heteroarylation of aryl sulfonium salts using alpha-amino alkyl radicals, which differs mechanistically from previously reported halogen-atom transfer (XAT). The new activation mode of aryl sulfonium salts can be carried out in the absence of light and photoredox catalysts, and is applicable to a wide range of heterocycles, showing potential for useful cross-coupling transformations.

Synthesis and evaluation of 3′-fluorinated 7-deazapurine nucleosides as antikinetoplastid agents

Jakob Bouton et al.

Summary: Kinetoplastid parasites rely on purine salvage for their purine demand. Evaluating the effect of introducing a 3'-fluoro substituent in 7-deazapurine nucleosides, several analogs with potent anti-parasitic activity were discovered, showing that a fluorine atom in the 3'-position is a promising modification for the discovery of anti-parasitic nucleosides.
