TADs enriched in histone H1.2 strongly overlap with the B compartment, inaccessible chromatin, and AT-rich Giemsa bands

Nuria Serna-Pujol et al.

Summary: The research found that G-positive bands are enriched in gene richness, active marks, and early replication, while G-negative bands are enriched in repressive marks, lamina-associated domains, and LINEs. Histone H1 variants distribute heterogeneously among G bands, and based on epigenetic features and H1 content, G bands can be organized in clusters useful for compartmentalizing the genome.

Unraveling linker histone interactions in nucleosomes

Fanfan Hao et al.

Summary: Significant progress has been made recently in defining the interactions of linker histones (H1 s) within nucleosomes, particularly in understanding the structures and interactions of the globular domains. However, more information is needed regarding the intrinsically disordered N-terminal and C-terminal domains.

Binding Dynamics of Disordered Linker Histone H1 with a Nucleosomal Particle

Hao Wu et al.

Summary: Linker histone H1 is crucial for chromatin packaging and gene expression, with dysregulation commonly seen in tumor cells. It alters chromatin condensation levels by binding to multiple sites on nucleosomal particles. The disordered domains of H1 enhance binding affinity and restrict dynamics, ultimately modulating nucleosome structure and dynamics.

Histone dynamics mediate DNA unwrapping and sliding in nucleosomes

Grigoriy A. Armeev et al.

Summary: Nucleosomes tightly wrap around 147 DNA base pairs with an octamer of histone proteins, and their dynamics enable DNA unwrapping and sliding. Molecular dynamics simulations revealed functional modes of nucleosome dynamics such as DNA breathing, unwrapping, twisting, and sliding. This study highlights the importance of nucleosomal DNA dynamics in chromatin conformational variability and the effects of chromatin modifications.

DNA sequence-dependent positioning of the linker histone in a nucleosome: A single-pair FRET study

Madhura De et al.

Summary: This study investigated the effect of linker DNA sequence on the binding of linker histones to nucleosomes. The results showed that linker DNA containing A-tract sequence consistently exhibited higher binding efficiency compared to pure-GC sequences. Furthermore, the study proposed a model to explain the binding mechanism of the globular domain of linker histones with different DNA sequences.
