Fatty Acids Can Induce the Formation of Proteinaceous Particles in Monoclonal Antibody Formulations

Summary: The presence of subvisible or visible particles in mAb formulations can cause problems in pharmaceutical development, especially concerning proteinaceous particles and their potential immunogenicity. This study explores the formation of protein particles and suggests that both interfaces and mechanical agitation as well as fatty acids, impurities or degradation products, can contribute to their formation. Specifically, oleic acid was found to be more effective than lauric acid in causing protein particles, and the presence of polysorbate mitigated the effect of fatty acids significantly.

The driving forces and rate of adsorption of a monoclonal antibody to the silicone rubber surface, as well as the influence of formulation parameters on particle concentration and size distribution, were investigated. It was found that formulation parameters, pH, and ionic strength, had an effect on the total amount of adsorbed protein and the formation of particles larger than 25 mm.
