Monitoring polysorbate hydrolysis in therapeutic proteins using an ultrasensitive extraction-free fatty acid quantitation method

Fatty Acids Can Induce the Formation of Proteinaceous Particles in Monoclonal Antibody Formulations

Summary: The presence of subvisible or visible particles in mAb formulations can cause problems in pharmaceutical development, especially concerning proteinaceous particles and their potential immunogenicity. This study explores the formation of protein particles and suggests that both interfaces and mechanical agitation as well as fatty acids, impurities or degradation products, can contribute to their formation. Specifically, oleic acid was found to be more effective than lauric acid in causing protein particles, and the presence of polysorbate mitigated the effect of fatty acids significantly.

The developed method demonstrates unique monoester peak profile of PS80 from different vendors, attributed by differences in relative abundance of the fatty acid monoesters. The method also revealed the different susceptibility of PS80 at different grades to auto-oxidation and hydrolysis.

Mechanistic understanding of metal-catalyzed oxidation of polysorbate 80 and monoclonal antibody in biotherapeutic formulations

Summary: This study evaluated the oxidation of polysorbate 80 (PS80) and a monoclonal antibody (mAb) under different conditions. The presence of iron accelerated the oxidation of PS80 and increased the oxidation of a specific solvent-exposed mAb methionine. PS80-AO was more sensitive to oxidative degradation than PS80-MC. The kinetics of PS80 oxidation was pH-dependent in the presence of histidine buffer. Increasing mAb concentration exhibited a protective effect against metal catalyzed PS80 and methionine oxidation.

4-Hydroxynonenal is An Oxidative Degradation Product of Polysorbate 80

Summary: This study evaluated whether HNE may be a pharmaceutically relevant degradation product of PS80 and established a new quantification method to detect its presence. The results confirmed HNE as a degradant in oxidatively stressed samples and showed that the antioxidant BHT can prevent its formation.
