A state of the art in analytical quality-by-design and perspectives in characterization of nano-enabled medicinal products

Thierry Bastogne et al.

Summary: Quality-by-Design (QbD) is a risk-based and proactive approach widely used in the pharmaceutical field for drug development. Analytical Quality by Design (AQbD) is the adaptation of QbD for analytical method development, aiming to optimize the accuracy and robustness of analysis results. However, there are challenges in understanding and implementing the AQbD approach in practice.

Asymmetric-flow field-flow fractionation for measuring particle size, drug loading and (in)stability of nanopharmaceuticals. The joint view of European Union Nanomedicine Characterization Laboratory and National Cancer Institute - Nanotechnology Characterization Laboratory

F. Caputo et al.

Summary: Asymmetric-flow field-flow fractionation (AF4) is a valuable tool for characterizing nanoparticle properties, such as size, polydispersity, drug loading, and stability. Through an international collaboration, a robust standard operating procedure (SOP) for measuring physical-chemical properties of nanopharmaceuticals using multi-detector AF4 (MD-AF4) has been developed. The joint experience of NCI-NCL and EUNCL demonstrates the ability to implement highly complex analytical techniques to meet regulatory standards for preclinical safety characterization of nanomedicines.
