Disruption of pancreatic stellate cell myofibroblast phenotype promotes pancreatic tumor invasion

Elizabeth R. Murray et al.

Summary: PKN2 plays a critical role in the differentiation of pancreatic stellate cells into myofibroblast-like cancer-associated fibroblasts. Loss of PKN2 leads to reduced proliferation and contractility of pancreatic stellate cells, as well as a switch from myofibroblast to inflammatory cancer-associated fibroblast phenotype. Furthermore, deletion of PKN2 promotes invasive cancer cell outgrowth and induces more locally invasive pancreatic tumors. The absence of PKN2 in the pancreatic stroma predicts poor outcome in pancreatic and other solid human cancers.

Composition, Spatial Characteristics, and Prognostic Significance of Myeloid Cell Infiltration in Pancreatic Cancer

Summary: The study revealed a diverse set of myeloid cells within the PDAC tumor microenvironment, which are distributed heterogeneously across patient tumors. Beyond cell density, the spatial locations of immune cells are also associated with patient outcomes, underscoring the potential role of spatially resolved myeloid cell subtypes as quantitative biomarkers for PDAC prognosis and therapy.

Spatially confined sub-tumor microenvironments in pancreatic cancer

Barbara T. Grunwald et al.

Summary: Through large-scale integration of histology-guided regional multiOMICs with clinical data and patient-derived preclinical models, we identified distinct subTMEs in the human pancreatic tumor microenvironment (TME) that are linked to tumor immunity, subtypes, differentiation, and treatment response. Reactive subTMEs rich in coordinated fibroblast communities were immune hot, while matrix-rich deserted subTMEs harbored fewer activated fibroblasts but showed chemoprotective features. These subTMEs originated in fibroblast differentiation trajectories and produced patient-specific heterogeneity tightly linked to malignant biology.

Pharmacologic Normalization of Pancreatic Cancer-Associated Fibroblast Secretome Impairs Prometastatic Cross-Talk With Macrophages

Remi Samain et al.

Summary: The study suggests that SOM230 has the potential to be an effective anti-metastatic therapy in PDA by regulating stromal activities, reducing inflammatory responses, and improving the immunosuppressive tumor microenvironment. This pharmacotherapy shows promise in reshaping the tumor environment and enhancing treatment outcomes for pancreatic cancer patients.

Leukocyte Heterogeneity in Pancreatic Ductal Adenocarcinoma: Phenotypic and Spatial Features Associated with Clinical Outcome

Summary: Researchers have established an immune atlas of PDAC through single-cell resolution profiling, identifying leukocyte features correlated with clinical outcomes, which can serve as a valuable reference for PDAC treatment. The data can aid in deeper understanding of the PDAC immune ecosystem, identifying additional therapeutic targets, and providing predictive biomarkers for therapeutic response and resistance monitoring.

Summary: This cross-sectional study projected cancer incidences and deaths in the United States up to 2040. The findings indicate significant changes in the most common cancers by 2040, with breast cancer projected to become the most common cancer and lung cancer remaining the leading cause of cancer-related death.

Single-cell analysis defines a pancreatic fibroblast lineage that supports anti-tumor immunity

Colin Hutton et al.

Summary: This study used mass cytometry to analyze the stromal composition in murine tissues and tumors, revealing extensive stromal heterogeneity and coordinated relationships between mesenchymal and immune cell subsets in pancreatic ductal adenocarcinoma. The research identified two stable and functionally distinct pancreatic fibroblast lineages marked by CD105 expression, with CD105-positive fibroblasts promoting tumor growth and CD105-negative fibroblasts suppressing tumors, dependent on adaptive immunity. These findings highlight the importance of mesenchymal and immune cell interactions in restricting tumor growth.

Tumor restriction by type I collagen opposes tumor- promoting effects of cancer-associated fibroblasts

Sonakshi Bhattacharjee et al.

Summary: This study reveals that cancer-associated fibroblasts (CAF) promote tumor growth by interacting with tumor cells through the secretion of different mediators. However, the overall tumor-promoting effects of CAF can be opposed by mechanical restriction from type I collagen. Therapeutic targeting of tumor-promoting CAF mediators while preserving type I collagen may shift the balance from tumor promotion to tumor restriction.
