Astrocytes Mediate Cholinergic Regulation of Adult Hippocampal Neurogenesis and Memory Through M1 Muscarinic Receptor

Summary: This study investigates the role of astrocytic CHRM1 in the regulation of adult hippocampal neurogenesis (AHN) and memory. The results show that genetic ablation of CHRM1 in astrocytes leads to defects in neural stem cell survival, neuronal differentiation, and maturation and integration of newborn neurons in the dentate gyrus. Astrocytic CHRM1-mediated modulation of AHN is mediated by BDNF signaling. Furthermore, CHRM1 ablation in astrocytes impairs contextual fear memory. These impairments in both AHN and memory are rescued by overexpression of astrocytic CHRM1 in the dentate gyrus.

PBM also reduces amyloid load, AMPA receptor endocytosis, dendrite injury, and inflammatory responses, rescuing memory deficits in APP/PS1 mice. Mechanistically, PBM activates ERK to inactivate JNK3 by phosphorylating and stabilizing MKP7, suggesting a potential therapeutic strategy for AD.
