Atomistic Model of Solute Transport across the Blood-Brain Barrier

Christian Jorgensen et al.

Summary: The study used atomistic MD simulations to visualize the translocation of solutes across a lipid bilayer representative of brain microvascular endothelial cells. Solutes were classified into three groups based on free-energy surface profiles, revealing distinct transport mechanisms. The simulations demonstrated the potential for MD simulations to provide new insight into brain penetration mechanisms and drug discovery approaches.

Prediction of the Blood-Brain Barrier (BBB) Permeability of Chemicals Based on Machine-Learning and Ensemble Methods

Lili Liu et al.

Summary: An ensemble learning model was developed to predict the blood-brain barrier (BBB) permeability of compounds using three machine-learning algorithms and nine molecular fingerprints, achieving high accuracy and sensitivity in cross-validation. The ensemble model outperformed most base classifiers and showed good accuracy in external validation, indicating its potential for early screening of central nervous system drugs.

Strategies for Structural Modification of Small Molecules to Improve Blood-Brain Barrier Penetration: A Recent Perspective

Baichen Xiong et al.

Summary: The blood-brain barrier poses challenges in delivering drugs to the brain, with small molecule drugs dominating current CNS medication. Medicinal chemists have been working on enhancing the penetration of small molecules by improving diffusion, reducing efflux, and activating carrier transporters, aiming to provide insights for future development of small molecule CNS drugs.

Predicting the Membrane Permeability of Fentanyl and Its Analogues by Molecular Dynamics Simulations

Christopher Faulkner et al.

Summary: This study used molecular dynamics simulations to investigate the permeation process of fentanyl and its analogues into phospholipid membranes, accurately predicting the permeability coefficients of these drug molecules. It was found that one specific phospholipid provided more accurate predictions than other commonly used lipids, which will be helpful for future studies in this area.
