Phase-selective staining of model and cell membranes, lipid droplets and lipoproteins with fluorescent solvatochromic pyrene probes

Jesus Sot et al.

Summary: The push-pull solvatochromic pyrene derivatives PA and PK have been successfully applied as polarity-sensitive probes in the study of model membrane vesicles, cells and purified human serum lipoproteins. These probes provide information about lipid order in biomembranes, with the advantage of being excitable by a standard 405 nm laser light. The results from the study showed the capability of PA and PK to report differently on liquid-disordered, liquid-ordered and gel phase bilayers.

Targeted Solvatochromic Fluorescent Probes for Imaging Lipid Order in Organelles under Oxidative and Mechanical Stress

Dmytro Danylchuk et al.

Summary: A series of solvatochromic probes have been designed for specific targeting of different organelles in live cells, allowing the detection of variations in lipid order and quantitative description of lipid order in each organelle. These probes revealed the characteristics of organelles under stress conditions, with mitochondria and lysosomes particularly sensitive to stress. The new probes provide a powerful toolbox for monitoring cellular response to physical and chemical stimuli at the level of individual organelle membranes.

LAURDAN since Weber: The Quest for Visualizing Membrane Heterogeneity

German Gunther et al.

Summary: Chemists studying membrane bilayer heterogeneity may encounter phase segregation in artificial bilayers and small sterol and sphingolipid-enriched dynamic domains in biological bilayers. Techniques using fluorescent probes and microscopy can assess membrane heterogeneity at different temporal and spatial levels, providing valuable information about the physical-chemical aspects of the membrane.

Emerging solvatochromic push-pull dyes for monitoring the lipid order of biomembranes in live cells

Yosuke Niko et al.

Summary: Solvatochromic dyes have emerged as a new class of fluorescent probes in lipid membranes field due to their ability to identify lipid organization in live cells by changing fluorescence color. The heterogeneous features of biomembranes caused by uneven lipid and cholesterol distribution in live cells can be studied using these dyes. Advanced solvatochromic dyes have been rapidly developed over the last decade, enabling applications in fluorescence microscopy and cellular studies.

Emerging solvatochromic push-pull dyes for monitoring the lipid order of biomembranes in live cells

Yosuke Niko et al.

Summary: Solvatochromic dyes have emerged as important fluorescent probes in the field of lipid membranes, allowing researchers to study the heterogeneous features of biomembranes by changing the color of their fluorescence. Recent developments in solvatochromic dyes, particularly those based on pyrene and Nile red, have shown promise for applications in cell biological and biophysical studies.
