PRE-driven protein NMR structures: an alternative approach in highly paramagnetic systems

A study utilized NMR to determine protein structure, finding that under favorable conditions, Paramagnetic Relaxation Enhancements (PREs) can efficiently complement and eventually replace Nuclear Overhauser Enhancements (NOEs) for structural characterization of small paramagnetic metalloproteins or de novo-designed metalloproteins.

A Chiral Lanthanide Tag for Stable and Rigid Attachment to Single Cysteine Residues in Proteins for NMR, EPR and Time-Resolved Luminescence Studies

Summary: The study introduces a new stable chiral Ln(III) tag, known as C12, for site-specific attachment to proteins, enabling protein probing through various spectroscopic techniques. The C12 tag exhibits enhanced reactivity towards selenocysteine, offering a pathway for selectively tagging selenocysteine in proteins containing cysteine residues. With Tb-III or Tm-III ions, the C12 tag can generate pseudocontact shifts in protein NMR spectra, providing a rigid tether between lanthanide and protein for interpretation of the shifts and distance measurements in double electron-electron resonance experiments. Additionally, the Tb-III complex upon reaction with cysteine or thiol compounds shows a significant enhancement in luminescence quantum yield, making it a sensitive luminescence probe for time-resolved FRET assays and enzyme reaction monitoring.

Fe-S cofactors in the SARS-CoV-2 RNA-dependent RNA polymerase are potential antiviral targets

Summary: Research has shown that the catalytic subunit nsp12 of the SARS-CoV-2 RdRp complex ligates two iron-sulfur metal cofactors which are essential for viral replication and interaction with the viral helicase. Oxidation of these clusters by TEMPOL inhibits RdRp activity and blocks SARS-CoV-2 replication in cell culture, indicating that these iron-sulfur clusters could be targeted for therapy of COVID-19.

Histidine orientation in artificial peroxidase regioisomers as determined by paramagnetic NMR shifts

Summary: Fe-Mimochrome VI*a is a synthetic peroxidase and peroxygenase with two different peptides covalently linked to deuteroheme. Shortening the distance between the distal peptide and heme allowed for the separation and characterization of two regioisomers, which differ in activity and axial-ligand orientation. These findings suggest that synthetic metalloenzymes could be useful for understanding the role of axial ligand orientation in peroxidase activity.
