SREBP1 promotes invasive phenotypes by upregulating CYR61/CTGF via the Hippo-YAP pathway

Chi-Yu Kuo et al.

Summary: Aberrant lipid metabolism plays a crucial role in providing energy, building materials, and oxidative stress resistance to cancer cells. This study found that high expression of SREBP1 in invasive thyroid cancer is associated with extrathyroidal extension, advanced disease stage, and shorter disease-specific survival. SREBP1 overexpression promotes cancer cell invasiveness through the upregulation of CYR61/CTGF via the Hippo-YAP pathway.

Thyroid Cancer Incidence and Mortality Trends in the United States: 2000-2018

Uchechukwu C. C. Megwalu et al.

Summary: Thyroid cancer incidence rates have declined since 2014, with similar declines noted for papillary thyroid carcinoma (PTC), localized disease, and tumors <= 1.0 cm, but not for tumors >2.0 cm. Incidence-based mortality rates continue to increase. These findings suggest changes in the management of thyroid nodules may have led to a decrease in the diagnosis of small indolent tumors, but not more advanced tumors.

Molecular diagnosis and targeted treatment of advanced follicular cell-derived thyroid cancer in the precision medicine era

Jaume Capdevila et al.

Summary: Most malignant thyroid tumours are initially treated with surgery or a combination of surgery and radioactive iodine therapy. However, in patients with metastatic disease, many tumours become refractory to radioactive iodine (RAI), and these patients require alternative treatments, such as locoregional therapies and/or systemic treatment with multikinase inhibitors. Targeted therapies based on genetic alterations have shown clinical efficacy in thyroid cancer, including NTRK gene fusions, RET and BRAF inhibitions. Effective testing strategies for these genetic alterations are crucial for identifying patients who could benefit from targeted treatments.

Characterization of TERT and BRAF copy number variation in papillary thyroid carcinoma: An analysis of the cancer genome atlas study

Brittany A. McKelvey et al.

Summary: This study focused on the role of TERT and BRAF copy number variations in papillary thyroid cancer (PTC), revealing that TERT amplifications are more common in later stage tumors and are frequently found in tumors with TERT mutations, while BRAF amplifications are more commonly associated with BRAF wildtype tumors and certain subtypes of PTC. Analyzing the differences in patterns of TERT and BRAF amplifications in relation to mutation status may contribute to a better understanding of cancer drivers in thyroid cancer.
