Smarca4 Inactivation Promotes Lineage- Specific Transformation and Early Metastatic Features in the Lung

Carla P. Concepcion et al.

Summary: The loss of Smarca4 results in increased sensitivity to malignant transformation and tumor progression in lung cells, leading to advanced dedifferentiated tumors and higher metastatic incidence. Smarca4-deficient tumors lack lung lineage transcription factor activities and resemble a metastatic cell state, suggesting a critical role of Smarca4 in regulating cellular transformation and metastasis during lung cancer evolution.

Repurposing Anticancer Drugs for the Treatment of Idiopathic Pulmonary Fibrosis and Antifibrotic Drugs for the Treatment of Cancer: State of the Art

Panagiotis Paliogiannis et al.

Summary: Idiopathic pulmonary fibrosis (IPF) and lung cancer share similarities in molecular, pathophysiological, and clinical aspects, with high mortality rates. Anti-fibrotic drugs like Nintedanib and Pirfenidone have potential for treating lung cancer, while novel targeted agents and immunotherapies may have anti-fibrotic properties. This suggests the possibility of using anti-fibrotic and anti-neoplastic drugs in combination for the treatment of these deadly diseases.

Categorization of lung mesenchymal cells in development and fibrosis

Xue Liu et al.

Summary: Pulmonary mesenchymal cells play critical roles in lung development and disease states in both mice and humans, showing high heterogeneity and subtype-specific transcriptomic differences which contribute to matrix gene expression in fibrosis. Comparative analysis reveals conserved transcriptomic signatures in orthologous subpopulations of murine and human lung mesenchymal cells, enhancing our understanding of mesenchymal cell diversity in lung development and disease conditions.

Structure-based classification predicts drug response in EGFR-mutant NSCLC

Jacqulyne P. Robichaux et al.

Summary: The study characterized the mutational landscape in 16,715 patients with EGFR-mutant NSCLC and established the structure-function relationship of EGFR mutations on drug sensitivity. EGFR mutations can be separated into four distinct subgroups based on sensitivity and structural changes, predicting patient outcomes following treatment with EGFR inhibitors better than traditional exon-based groups. This structure-based approach delineates functional groups of EGFR mutations that can effectively guide treatment choices and suggest potential improvement in prediction of drug sensitivity to targeted therapies in oncogenes with diverse mutations.
