GSDMB is increased in IBD and regulates epithelial restitution/repair independent of pyroptosis

Nitish Rana et al.

Summary: Increased GSDMB in inflammatory bowel disease affects the restoration of epithelial barrier function and resolution of inflammation. Mechanistic experiments reveal that GSDMB deficiency impairs the increased proliferation and migration of epithelial cells during in vitro wound closure.

Gasdermin-B Pro-Tumor Function in Novel Knock-in Mouse Models Depends on the in vivo Biological Context

David Sarrio et al.

Summary: Gasdermin-B (GSDMB) plays complex roles in inflammatory diseases and cancer. In this study, a knock-in mouse model expressing human GSDMB2 was generated to investigate its functions in cancer. The results showed that GSDMB2 has a pro-tumor effect in breast cancer when in concert with the HER2 oncogene, but has no overall intrinsic oncogenic potential in genetically modified mice. These findings provide insights into the specific biological contexts in which GSDMB2 functions as a pro-tumor factor.

Channelling inflammation: gasdermins in physiology and disease

Xing Liu et al.

Summary: Gasdermins have been identified as mediators of inflammatory cell death known as pyroptosis, forming cell membrane pores upon activation to release pro-inflammatory cytokines. They play important roles in autoimmune and inflammatory diseases, infectious diseases, deafness, and cancer, with potential for novel therapeutic avenues.

Pathogenic ubiquitination of GSDMB inhibits NK cell bactericidal functions

Justin M. Hansen et al.

Summary: GSDMB is targeted and destroyed by the IpaH7.8 effector protein secreted by Shigella flexneri, protecting the bacterium from natural killer cells. Unlike other gasdermin family members, GSDMB exhibits direct microbiocidal activity by recognizing phospholipids found on Gram-negative bacterial membranes.

HER2-positive breast cancer and tyrosine kinase inhibitors: the time is now

Ilana Schlam et al.

Summary: HER2-positive breast cancer accounts for a significant proportion of cases, with several targeted therapies available including TKIs which have shown promising responses in both early and advanced settings. In patients with central nervous system involvement, TKIs have shown efficacy, which is important for those with limited treatment options and poor prognosis in this setting.

USP24-GSDMB complex promotes bladder cancer proliferation via activation of the STAT3 pathway

Haiqing He et al.

Summary: The study demonstrated that GSDMB promotes bladder cancer progression and interacts with STAT3 to activate signaling pathways in tumor cells. Moreover, USP24 stabilizes GSDMB to activate STAT3 signaling in bladder cancer cells.

Nlp promotes autophagy through facilitating the interaction of Rab7 and FYCO1

Wenchang Xiao et al.

Summary: Autophagy is a crucial pathway for maintaining intracellular homeostasis and preventing malignant transformation. The study reveals that Nlp, a centrosomal protein, is a novel regulator of autophagy that enhances the interaction between Rab7 and FYCO1, facilitating autophagolysosome formation and accelerating autophagic flux. Deficiency of Nlp leads to hepatic autophagic defects and increased incidence of liver cancer in mice.
