Ultrasound as a tool for polymorph control and high yield in flow crystallization

Mohammed Noorul Hussain et al.

Summary: The study showed that using ultrasound combined with seed guidance effectively increased the yield and purity of form-I of o-aminobenzoic acid. Compared to silent conditions in flow crystallization experiments, ultrasound improved seed dispersion and reduced wall nucleation, resulting in higher quality pure product.

Progress and Principle of Drug Nanocrystals for Tumor Targeted Delivery

Meng Bai et al.

Summary: Drug nanocrystals refer to drugs existing as nanoscale crystal structures, widely used to enhance solubility and absorption of poorly aqueous soluble drugs or prolong drug circulation. To address the poor targetability, drug nanocrystals for tumor targeted delivery have been developed, with various surface modifications introduced for passive, active, and stimuli-responsive targeting mechanisms.

Concomitant Polymorphism and Nucleation Control of DL-Methionine Through Antisolvent Crystallization

Manivel Suresh et al.

Summary: In this study, researchers discovered the concomitant polymorphism of the sulfur-containing essential amino acid DL-methionine and successfully separated the alpha- and beta-polymorphic forms through antisolvent crystallization with ethanol. The addition of varying volume fractions of ethanol into the solution affected the solubility of DL-methionine and led to different levels of supersaturation, resulting in the formation of stable beta-polymorphs, mixture of alpha- and beta-polymorphs, and metastable alpha-polymorphs. The distinct needle-shaped alpha-polymorphs and platy-shaped beta-polymorphs were confirmed through optical microscopic and X-ray diffraction analyses.

Polymorphic Control of α and β dl-Methionine through Swift Cooling Crystallization Process

Manivel Suresh et al.

Summary: Polymorphic control and crystal nucleation of alpha and beta polymorphs of dl-methionine, an essential amino acid, can be achieved from saturated aqueous solution through swift cooling crystallization process, by generating a wide range of supersaturation (0.02 < sigma < 2.19) in the temperature range of 323-274 K. The type of nucleation is highly dependent on the level of supersaturation: lower supersaturation yields the stable beta form, while higher supersaturation results in the metastable alpha form. In situ optical microscopy and X-ray diffraction analyses confirm the morphological and structural characteristics of the grown polymorphs.

Design of Spherical Crystallization of Active Pharmaceutical Ingredients via a Highly Efficient Strategy: From Screening to Preparation

Yiming Ma et al.

Summary: This work aims to develop a highly efficient spherical crystallization process based on liquid-liquid phase separation (LLPS) from screening to preparation stage. Machine learning models were used to predict LLPS for an API, with water and ethanol chosen as solvents to reduce toxicity and energy consumption.
