Size Exclusion and Ion Exchange Chromatographic Hardware Modified with a Hydrophilic Hybrid Surface

Szabolcs Fekete et al.

Summary: In order to address the challenges in analyzing certain biomolecules using liquid chromatography, researchers have developed a new hydrophilically modified hybrid surface (h-HST) for size exclusion chromatography and anion exchange separations. By incorporating additional oxygen and carbon atoms onto an ethylene bridge hybrid siloxane polymer, this surface reduces non-specific adsorption and undesired secondary interactions, resulting in improved separations of proteins and nucleic acids.

Influence of metals in the column or instrument on performance in hydrophilic interaction liquid chromatography

David McCalley

Summary: A method for measuring the relative contribution of extracolumn and column effects to the detrimental interactions in the HPLC system is proposed. The study focused on HILIC separations and found that using a deactivated HILIC column treated by a vapor deposition procedure and adding metal complexing agents improved the separation efficiency by reducing the interactions between metals and solutes.

Current state-of-the-art of separation methods used in LC-MS based metabolomics and lipidomics

Eva-Maria Harrieder et al.

Summary: Metabolomics and lipidomics involve the large-scale analysis of metabolites, requiring advanced analytical methods. Liquid Chromatography-Mass Spectrometry (LC-MS) is a commonly used technique that provides different selectivities in separation and high sensitivity in detection. Due to the huge chemical diversity, there is no single analysis method that can cover the entire range of metabolites or lipids, leading to the use of different separation methods. This review explores the current use of LC-MS in metabolomics and lipidomics using data from public databases, and highlights potential future trends and improvements needed.

Artemis Lioupi et al.

Summary: In this study, metabolic phenotyping of mouse liver extracts was conducted using a novel zwitterionic HILIC UHPLC column. The column exhibited efficient chromatographic separations and enabled the analysis of approximately 80 hydrophilic metabolites. Comprehensive metabolic signatures of the tissue extracts were obtained, providing insights into the metabolic phenotypes.

Internal calibration as an emerging approach for endogenous analyte quantification: Application to steroids

Gioele Visconti et al.

Summary: In this study, an innovative internal calibration approach using stable isotope-labeled standard version of endogenous compound was developed to improve the quantitative performance of mass spectrometry methods. The results showed that the response factors obtained on different LC-MS platforms were consistent and the quantitative results in human serum samples were similar to those obtained with conventional quantification approach based on external calibration. Furthermore, it was demonstrated that the naturally occurring isotopes of SIL could be used to increase the accuracy for analytes with low concentrations.
